宫颈癌组织miR-219a表达水平与患者肿瘤侵袭转移及预后的相关性

摘　　要：目的研究宫颈癌组织微小RNA-219a(miR-219a)的表达水平与患者肿瘤侵袭转移及预后之间的关系。方法选取2014年9月—2015年9月收治的86例宫颈癌肿瘤组织和25例正常对照的宫颈组织样本,用qRT-PCR检测组织中miR-219a的表达水平。结合宫颈癌患者临床资料,分析miR-219a与患者的临床病理特征及3年生存率之间的关系。结果宫颈癌肿瘤组织miR-219a相对表达量低于正常宫颈组织,miR-219a低表达率显著高于正常宫颈组织(P<0.01)。miR-219a表达水平与宫颈癌分化程度、国际妇产科联合会(FIGO)分期、淋巴转移、间质浸润深度、阴道壁累及有相关性(P<0.05),与年龄、病理分型、肿瘤大小及宫旁组织累及无关(P>0.05)。miR-219a低表达患者的3年总生存率和3年无病生存率均显著低于miR-219a高表达患者(P<0.01)。结论宫颈癌肿瘤组织中miR-219a表达下调,并且可能与宫颈癌患者的分化程度、FIGO分期、淋巴转移、间质浸润深度、阴道壁累及相关,同时miR-219a低表达可能提示患者3年生存率降低。Objective To study correlations between expression of microRNA-219a(miR-219a)and tumor invasion,metastasis and prognosis in patients with cervical cancer.Methods A total of 86 tumor tissue cases of cervical cancer and 25 cervical tissue samples as normal control during September 2014 and September 2015 were selected,and expression of miR-219a in tissues were detected by quantitative reverse transcription-polymerase chain reaction(qRT-PCR).Correlations between miR-219a with patients'clinical pathological features and 3-year survival period were analyzed combined with clinical data of patients with cervical cancer.Results Relative expression of miR-219a in cervical cancer tissues was significantly lower,and low expression rate of miR-219a was significantly higher than those in normal cervical tissues(P<0.01).Expression of miR-219a was correlated with differentiation degree of cervical cancer,federation international of gynecology and obstetrics(FIGO)staging,lymphatic metastasis,interstitial infiltration depth and vaginal wall involvement(P<0.05),but it was not correlation with age,pathological classification,tumor size and involvement of parametrial tissues(P>0.05).The 3-year overall survival rate and 3-year disease-free survival rate of patients with low expressed miR-219a were significantly lower than those in patients with high expressed miR-219a(P<0.01).Conclusion Down-regulated expression of miR-219a in cervical cancer tissue may be correlated with cervical cancer differentiation,FIGO staging,pelvic lymphatic metastasis,interstitial infiltration depth and vaginal wall involvement,and low expressed miR-219a may indicate decrease of 3-year survival rate of patients.
