摘 要:目的通过观察瑞舒伐他汀对急性脑缺血再灌注损伤大鼠Bax、Bcl-2、脑含水量的影响,探讨其脑保护作用机制。方法将72只SD大鼠随机分为3组:A组为假模型组,B组为模型组,C组为干预组。采用改良的Zea Longa线栓法制作大鼠大脑中动脉缺血再灌注(MCAO)模型,免疫组化法观测大鼠海马区Bax、Bcl-2改变,干湿质量法测定脑含水量变化,神经功能评分采用Zea Longa 5分制标准评分,脑梗死容积百分比采用2,3,5苯四氮唑(tetrazolium chloride,TTC)染色测定。结果A组各不同时间点大鼠Bax、Bcl-2的激活、脑含水量、神经功能评分及脑梗死容积百分比无明显改变。B、C组大鼠各不同时间点Bax、Bcl-2的激活、脑含水量、神经功能评分及脑梗死容积百分比与A组相比较,有显著性差异(P<0.05)。24 h及72 h时脑含水量、脑梗死容积百分比C组与B组相比,差异不明显(P>0.05),7 d时C组与B组相比,降低明显(P<0.05)。结论瑞舒伐他汀能明显抑制急性脑缺血再灌注大鼠神经元过度凋亡,减轻脑水肿,降低脑梗死容积,具有脑保护作用。神经元数目增多,神经元变性程度及空泡现象明显减轻。Objective To observe the effect of rosuvastatin on Bax,Bcl-2 and brain water content in rats with acute cerebral ischemia-reperfusion injury,and to explore the mechanism of brain protection.Methods 72 SD rats were randomly divided into 3 groups:A group was a sham model group,B group was a model group,and C group was an intervention group.A rat model of middle cerebral artery ischemia-reperfusion(MCAO)was established using a modified Zea Longa method.The changes of Bax and Bcl-2 in the hippocampus of rats were observed by immunohistochemistry.The changes in brain water content were measured by dry-wet weight method.The functional score was based on the Zea Longa 5 score.Percentage of cerebral infarct volume was measured with 2,3,5 tetrazolium chloride(TCC)staining.Results The activation of Bax,Bcl-2,brain water content,neurological function score and the percentage of cerebral infarct volume in rats of group A were not significantly changed at different time points.The activation of rat Bax,Bcl-2,brain water content,neurological function score and percentage of cerebral infarct at different time points in groups B and C were significantly different from those of group A(P<0.05).There was no significant difference in brain water content and percentage of cerebral infarct volume between group C and group B at 24 h and 72 h(P>0.05).On days 7 and 14 d,C group was significantly lower than that in group B(P<0.05).Conclusion Rosuvastatin can significantly inhibit neuronal apoptosis,reduce brain edema,and reduce cerebral infarct volume in rats with acute cerebral ischemia-reperfusion.
