摘  要:目的:探讨海藻类功能食品对肠道微生态失调小鼠的菌群调整及对非特异性免疫的调节作用,以进一步开发利用海洋藻类资源。方法:将SPF级昆明种小鼠随机分为正常对照组和干预组,干预组首先采用大剂量混合抗生素灌胃建立小鼠肠道微生态失调模型,再根据后期干预方式的不同分为:模型组、自然恢复组(正常喂养)和治疗组(海藻糖类配方鼠粮喂养);收集各组小鼠新鲜粪便,利用PCR-DGGE方法检测肠道菌群结构变化,获得细菌群落指纹图谱并进行分析,同时切取差异显著条带进行序列分析;酶联免疫吸附实验检测血清中IL-2、TNF-α生成水平,进而初步评价海藻类食品对小鼠非特异性免疫的影响。结果:抗生素灌胃后导致小鼠肠道菌群失调,相关免疫细胞因子水平下降,模型组小鼠体内Barnesiella intestinihominis等抑制机体免疫力的菌群量明显高于正常及治疗组,治疗组小鼠乳酸杆菌等有益菌及肠道菌群多样性恢复较好且小鼠血清细胞因子浓度明显升高。结论:海藻类功能食品能够帮助恢复肠道微生态平衡,增强实验小鼠免疫功能。Objective:To investigate the intestinal microflora and nonspecific immunity effect of algal functional food in mice with intestinal flora imbalance induced by multiple-antibiotics,to further develop and utilize the resources of marine algae.Methods:Total of Kunming mice were randomly divided into normal control group,model group,natural recovery group(normal feeding)and treatment group(functional food feeding).The intestinal microbe imbalance mice were established by intragastric administration of multiple-high dose antibiotics.The fresh feces of each mouse were collected and the fingerprints of the intestinal flora were obtained by PCR-DGGE and analysed by related software.Significantly different bands were cut off to do further sequencing analysis.The production level of serum cytokines IL-2 and TNF-αwere detected by enzyme-linked immunosorbent assay and to evaluate the effect of algal functional food on nonspecific immunity in mice.Results:The mice intestinal microbiota were disordered after being fed with antibiotics and the level of related cytokines dropped.The amount of Barnesiella intestinihominis in model group mice fecal which act as an important immune-inhibiting bacteria was obviously higher than that of normal and treatment group.After treatment fecal lactobacillus bacteria and intestinal flora diversity were all better recovered,and serum concentrations of cytokines in mice increased significantly.Conclusion:Algal functional food is beneficial to the recovery of lower immunity and reconstruction of intestinal microbiota in mice induced by antibiotics.
