摘  要:目的 探讨转化生长因子β活化的长链非编码RNA ATB(lncRNA ATB)在前列腺癌中的表达,以及通过调控miR-107/STAM结合蛋白水平1(STAMBPL1)通路在前列腺癌PC3细胞中对增殖、转移和侵袭的影响。方法 反转录PCR(RT-PCR)法检测58例前列腺癌组织及相应癌旁正常组织临床样本中lncRNA ATB mRNA的表达水平。siRNA干扰PC3细胞lncRNA ATB基因的表达,四甲基偶氮唑蓝(MTT)实验、划痕实验和Transwell实验分别观察PC3细胞增殖、迁移和侵袭能力的改变;采用RT-PCR和Western blot法分别测定miR-107mRNA及STAMBPL1mRNA、蛋白的表达变化。结果 与癌旁正常组织比较,lncRNA ATB mRNA在前列腺癌组织中表达水平明显增加(P<0.05)。沉默lncRNA ATB基因的PC3细胞较正常PC3细胞可显著减少细胞中增殖、迁移和侵袭能力,miR-107mRNA表达水平增高,STAMBPL1mRNA和蛋白表达水平降低(P<0.05)。结论 LncRNA ATB在前列腺癌中高表达,其可能通过调控miR-107/STAMBPL1通路改变前列腺癌PC3细胞增殖、转移和侵袭的能力。Objective To observe the expression of long-chain non-coding RNA activated by transforming growth factorβ(lncRNA ATB)in prostate cancer and its effect on proliferation,metastasis and invasion of prostate cancer PC3 cells by regulating miR-107/STAMBPL1 pathway.Methods RT-PCR method was used to measure the expression level of lncRNA ATB in 58 cases of prostate cancer tissues and adjacent normal tissues.siRNA interfered with the expression of lcnRNA ATB gene,MTT assay,cell scratch and transwell experiment were used to observe the effects of lncRNA ATB on cell proliferation,migration and invasion of PC3 cell.The mRNA and Protein expression levels of miR-107 and STAMBPL1 were measured by RT-PCR and Western blot,respectively.Results Compared with adjacent normal tissues,the expression level of lncRNA ATB in prostate cancer tissue was significantly increased(P<0.05);silencing of lncRNA ATB significantly reduced the proliferation,migration and invasion in prostate cancer PC3 cells,increased the expression level of miR-107 mRNA,and decreased the expression levels of STAMBPL1 mRNA and protein(P<0.05).Conclusion LncRNA ATB is highly expressed in prostate cancer,which may alter the proliferation,invasion and metastasis of prostate cancer PC3 cells by regulating miR-107/STAMBPL1 signaling pathway.
