PPARγ和P16在宫颈癌和宫颈鳞状上皮内病变患者宫颈组织中的表达及其与HPV感染的关系

摘　　要：【目的】探讨过氧化物酶体增殖物激活受体γ(PPARγ)和P16蛋白在宫颈鳞状细胞癌(SCC)和鳞状上皮内病变(SIL)患者宫颈组织中的表达及其与人乳头状瘤病毒(HPV)感染的关系。【方法】采用免疫组化MAXVisionTM法检测17例正常宫颈上皮(NCE)者和52例SIL及39例SCC患者宫颈组织中STAT3、P16蛋白表达,采用反向斑点杂交法检测HPV感染情况,分析PPARγ、P16的表达与HPV感染的相关性。【结果】PPARγ、P16在NCE组、SIL组及SCC组中的阳性率呈逐渐增高趋势,组间比较有统计学意义(P<0.05)。HPV在NCE组、SIL组、SCC组中的感染率分别为29.41%(5/17)、76.92%(40/52)、97.44%(38/39),也呈逐渐增高趋势,组间比较有统计学意义(P<0.05)。PPARγ、P16的表达均与HPV感染呈正相关(P<0.05)。【结论】PPARγ、P16在SCC组织中呈高表达,与HPV感染率呈正相关,其可能在SCC的发生过程中起重要作用。【Objective】To investigate the expression of peroxisome proliferator activated receptor gamma(PPARγ)and P16 protein in cervical squamous cell carcinoma(SCC)and cervical intraepithelial lesion(SIL),and possible correlation with HPV infection.【Methods】Expressions of PARγand P16 in 17 cases of normal cervical epithelium(NCE),52 cases of SIL and 39 cases of SCC were measured by immunohistochemical MAXVisionTM method.HPV infection was detected by reverse dot blot hybridization method.The correlation between the expression of PPARγand P16 with HPV infection was analyzed.【Results】The positive expression rate of PPARγand P16 from NCE to SIL and SCC showed a gradual increasing trend;the differences among groups were statistically significant(P<0.05).The infection rate of HPV in NCE,SIL and SCC was 29.41%(5/17),76.92%(40/52)and 97.44%(38/39),respectively,which also showed a gradual increasing trend.The difference between groups was statistically significant(P<0.05).The expressions of PPARγand P16 were positively correlated with HPV infection(P<0.05).【Conclusion】The expressions of PPARγand P16 are highly expressed in cervical SCC tissues.PPARγand P16 expressions are positively correlated with HPV infection,which play an important role in the development of cervical carcinoma.
