摘  要:为探索大孔吸附树脂纯化桔梗茎总黄酮的工艺条件及其体外抗氧化活性,以总黄酮的比吸附量、吸附率及比洗脱量、洗脱率为考察指标,采用单因素试验对桔梗茎总黄酮的纯化工艺进行考察;采用分光光度法探索了桔梗茎总黄酮对DPPH·、亚硝酸盐、ABTS阳离子自由基、·OH的清除活性。结果表明,桔梗茎总黄酮纯化的较佳工艺条件为:桔梗茎浸提液采用石油醚脱脂,控制其总黄酮质量浓度1.1 mg/mL,桔梗茎浸提液的上柱体积与树脂质量比6∶1(mL/mg),吸附流速控制1.0 mL/min,洗脱液为70%乙醇水溶液,洗脱流速控制2.0 mL/min,洗脱液体积与树脂质量比7∶1(mL/mg)。在此纯化条件下,LX-36型树脂对桔梗茎总黄酮的比吸附量平均为5.933 mg/g、吸附率平均为89.89%,比洗脱量平均为5.627 mg/g、洗脱率平均为94.83%,干浸膏中总黄酮含量由6.35%提高到40%以上。研究结果表明,桔梗茎总黄酮对DPPH·、亚硝酸盐、ABTS阳离子自由基、·OH均具有清除活性,而且均随着其质量浓度的增加,清除活性均逐渐增强。该结果为桔梗茎的进一步开发利用提供了依据。To explore the purification condition of total flavonoids from stems of Platycodon grandiflorum by macroporous resin and its antioxidant activity in vitro,the purification process of total flavonoids was optimized by single factor test using specific adsorbance,adsorption ratio,specific desorpted quantity and desorption ratio as investigation indexes.The scavenging activity of total flavonoids from Platycodon grandiflorum stems on DPPH·,NaNO2 and ABTS cation radicals,·OH was studied by spectrophotometry.The results showed that,the optimum purification conditions of total flavonoids in Platycodon grandiflorum stems were as follows,the stems extract was degreased with petroleum ether,the total flavonoids concentration 1.1 mg/mL,the ratio of extracted sample loading volume to resin mass 6∶1(mL/mg),the adsorption velocity 1.0 mL/min,the eluent 70%ethanol solution,the desorption velocity 2.0 mL/min,and the ratio of the eluent volume to resin mass 7∶1(mL/mg).Under these conditions,the average specific adsorbance,adsorption rate,specific desorpted quantity and desorption rate of LX-36 resin for total flavonoids were 5.933 mg/g,89.89%,5.627 mg/g and 94.83%,respectively.The content of total flavonoids in the dry extract could increase from 6.35%to over 40%.The antioxidant activity results showed that,total flavonoids from Platycodon grandiflorum stems had strong scavenging activity to DPPH·,NaNO2 and ABTS cation radicals and·OH,and the scavenging activity enhanced gradually with the increase of mass concentration of total flavonoids.The results provided basis for further development and utilization of Platycodon grandiflorum stems.
