摘　　要：目的:人cAMP依赖型蛋白激酶B抑制剂(cAMP-dependent protein kinase inhibitor-β,PKIB)表达对胰腺癌细胞增殖与侵袭的影响。方法:利用Lipofectamine TM 2000转染PKIB过表达和沉默基因至PANC-1胰腺癌细胞,应用实时定量聚合酶链反应(RT-PCR)法与蛋白质印迹法(Western blot)检测转染细胞中PKIB mRNA与蛋白的表达。应用噻唑蓝(MTT)比色法和结晶紫实验检测胰腺癌细胞活力和增殖能力,应用Transwell实验测定胰腺癌细胞的侵袭能力。结果:胰腺癌细胞的PKIB表达高于正常胰腺导管上皮细胞(P<0.05)。PKIB过表达增强细胞活力(P<0.05),促进细胞增殖,使胰腺癌细胞侵袭能力增强(P<0.05),而沉默PKIB表达对胰腺癌细胞的生物学效应恰好相反(P<0.05)。结论:PKIB的表达程度可影响胰腺癌细胞的增殖和侵袭,为进一步探讨胰腺癌发病机制及治疗提供有力证据。Objective:To explore the relationship between PKIB(cAMP-dependent protein kinase inhibitor-β)expression and the proliferation and invasion of human pancreatic cancer.Methods:PKIB and shRNA-PKIB were transfected into PANC-1 cells mediated by Lipofectamine TM 2000.Expression of PKIB mRNA and protein were respectively measured by real time polymerase chain reaction(RT-PCR)and Western blot.The cell viability and proliferation were measured by MTT assay and crystal violet assay.The cell invasion was assessed by Transwell assay.Results:We found that the expression of PKIB was up-regulated in pancreatic cancer tissue compared with the normal pancreatic tissue(P<0.05).Moreover,the overexpression of PKIB promotes the proliferation and the invasion cascade of pancreatic cancer cells(P<0.05).While silencing the PKIB gene expression with PKIB shRNA exerted the anti-effects(P<0.05).Conclusion:PKIB expression alteration effects the proliferation and invasion cascade of pancreatic cancer cells,which may be an important potential therapeutic target for pancreatic cancer.
