逍遥散对慢性轻度不可预见应激抑郁大鼠行为学及食欲调节因子的影响

摘　　要：目的:探讨逍遥散对慢性轻度不可预见应激抑郁大鼠的作用机理。方法:建立慢性轻度不可预见应激抑郁大鼠模型,随机分为模型组与逍遥散组各15只,另选10只正常大鼠作为对照组。逍遥散进行4周干预后,通过新环境进食压抑实验,观察各组大鼠摄食行为,ELISA法检测各组血清多巴胺(DA)、食欲素-A(orexin A)与瘦素(leptin)的变化并进行比较。结果:与正常组比较,模型组大鼠体质量、进食时间、进食量、血清DA、leptin和orexin A水平均显著降低,进食潜伏期明显延长。与模型组比较,逍遥散组大鼠体质量明显增加,血清DA与orexin A含量显著上升。结论:逍遥散对抑郁大鼠的体质量与食欲均有较好的治疗作用,可能与其调节orexin、leptin的平衡协调进而激活DA的分泌,发挥其在奖赏过程中的保护作用有关。Objective:To study the mechanisms of Xiaoyao Poweker treating rat of depression model.Methods:Rat models of chronic mild unpredictable stress depression were established and divided into 2 groups:the normal group(n=15)and the Xiaoyao Poweker group(n=15).After 4 weeks’treatment of Xiaoyao Poweker,the feeding behavior of rats in each group was observed by novelty suppressed feeding test,then the serum DA,leptin and orexin A levels were detected by Elisa test.Results:Compared with the normal group,the body weight,feeding time,food intake,serum DA,leptin and orexin A levels in the model group were significantly decreased,and the latency of feeding was significantly prolonged.Compared with the model group,the weight of rats in Xiaoyao Poweber group increased significantly,and the contents of serum DA and orexin A increased significantly.Conclusion:Xiaoyao Poweber has therapeutic effects on body weight and appetite of depressive rats,maybe it plays a protective role in the reward process by regulating the balance of orexin and leptin,thus activating the secretion of DA.
