摘 要:目的:观察MicroRNA-195对子宫内膜癌细胞增殖、迁移与侵袭能力的影响。方法:取对数生长期人子宫内膜癌细胞系Ishikawa,分别转染MicroRNA-195模拟物(MicroRNA-195过表达组)、MicroRNA-195抑制物(MicroRNA-195低表达组)及阴性对照质粒MicroRNA-NC(空白质粒组)。转染48h后,qRT-PCR法检测细胞中MicroRNA-195表达,CCK-8法检测细胞增殖,Transwell小室实验检测细胞迁移与侵袭能力,Western blot法检测Akt、p-Akt蛋白表达。将转染MicroRNA-195模拟物、MicroRNA-195抑制物及阴性对照质粒MicroRNA-NC的人子宫内膜癌细胞系Ishikawa分别注射至裸鼠皮下,每天检测肿瘤体积,实验周期为3周。结果:MicroRNA-195过表达可抑制人子宫内膜癌细胞系Ishikawa的增殖、迁移与侵袭能力(P<0.05),提高p-Akt蛋白表达(P<0.05);MicroRNA-195低表达可促进人子宫内膜癌细胞Ishikawa的增殖、迁移与侵袭能力(P<0.05),降低p-Akt蛋白表达(P<0.05)。MicroRNA-195过表达可抑制裸鼠体内人子宫内膜癌细胞Ishikawa的增殖(P<0.05),MicroRNA-195低表达可促进裸鼠体内人子宫内膜癌细胞Ishikawa的增殖(P<0.05)。结论:MicroRNA-195可抑制子宫内膜癌细胞Ishikawa的增殖、迁移与侵袭能力,其作用机制可能与激活PI3K/Akt信号通路有关。Objective:To observe the effect of microRNA-195 transfection on the proliferation,migration and invasion of endometrial cancer cells.Method:Human endometrial cancer cell line Ishikawa during the logarithmic growth period was transfected in three groups,MicroRNA-195 mimic(MicroRNA-195 over-expression group),MicroRNA-195 inhibitors(MicroRNA-195 low expression group),and negative control plasmid MicroRNA-NC(blank plasmid group).After transfection of 48 h,qRT-PCR was used to detect the expression of MicroRNA-195 in cells,CCK-8 was used to detect cell proliferation,Transwell small chamber experiment was used to detect cell migration and invasion ability,and Western blot was used to detect the Akt and p-Akt protein expressions.Ishikawa,a human endometrial cancer cell line transfected with microRNA-195 mimic,microRNA-195 inhibitor and negative control plasmid MicroRNA-NC,was subcutaneously injected into nude mice.Tumor volume was measured daily for 3 weeks.Result:MicroRNA-195 mimic and microRNA-195 inhibitor were successfully transfected into human endometrial cancer cell Ishikawa.Overexpression of microRNA-195 inhibited the proliferation,migration and invasion of human endometrial cancer cell line Ishikawa(P<0.05),and increased the expression of p-Akt protein(P<0.05).Low expression of microRNA-195 could promote the proliferation,migration and invasion of human endometrial cancer cell line Ishikawa(P<0.05),and decrease the expression of p-Akt protein(P<0.05).Overexpression of microRNA-195 inhibited the proliferation of human endometrial cancer cell Ishikawa in nude mice(P<0.05).Low expression of microRNA-195 promoted the proliferation of human endometrial cancer cell Ishikawa in nude mice(P<0.05).Conclusion:MicroRNA-195 can inhibit the proliferation,migration and invasion of endometrial cancer cell line Ishikawa,and its mechanism may be related to the activation of PI3 K/Akt signaling pathway.
