摘 要:采用不同浓度的秋水仙素、重金属溶液及混合液作为诱变剂,在25℃条件下分别培养大蒜根尖24 h和36 h.通过计算微核率及染色体加倍率,找出既能诱导产生微核,又能诱导产生多倍体细胞的最佳时间及诱变剂.实验表明,0.1%秋水仙素处理液可以同时产生微核和多倍体,且在处理36 h时多倍率最高,同时具有较高的微核率.筛选出0.1%秋水仙素溶液作为诱变剂,处理时间为36 h的效果最好,是作为一次设计同时完成诱导大蒜根尖产生微核和多倍体的最佳实验方法.In this paper different concentrations of colchicine,heavy metal solution and its mixture were used as mutagens.The root tip of garlic was cultured at 25℃for 24 h and 36 h.The micronucleus rate and chromosome doubling rate were calculated to find out the optimal time to induce the production of micronuclei and the induction of polyploid cells and the type of mutagen.Experiments show that 0.1%colchicine treatment can produce micronuclei and polyploid at the same time.When treated for 36 h the highest rate of magnification is gained,while having a higher micronucleus rate.Thus it is concluded that the best experimental method for inducing micronuclei and polyploidy at the root tip of garlic in one design at the same time is taking 0.1%colchicine solution as a mutagen with the treatment time of 36 h.
