摘 要:目的探讨绞股蓝皂甙(Gyp)对人结肠癌细胞增殖、凋亡的影响以及其可能的作用机制。方法体外培养人结肠癌细胞SW480,并分为空白对照组(Control)、低浓度Gyp组(50μmol/L)、中浓度Gyp组(100μmol/L)和高浓度Gyp组(200μmol/L)。采用克隆形成实验观察细胞生长情况,MTT法观察细胞增殖情况,流式细胞仪检测细胞凋亡情况,Western blot检测增殖凋亡蛋白及TGF-β/Smad信号通路相关蛋白表达情况。结果四组间细胞克隆形成率比较,差异有统计学意义(P <0.05),细胞克隆形成率随Gyp浓度的增加而降低。四组间24 h、48 h细胞抑制率以及四组间凋亡率比较,差异均有统计学意义(均P <0.05),细胞抑制率、凋亡率随Gyp浓度的增加而升高。四组间Ki67蛋白、Bcl-2蛋白表达水平差异均有统计学意义(均P <0.05),两者表达随Gyp浓度的增加而减少。四组间Caspase-3、Bax蛋白表达水平比较,差异均有统计学意义(均P <0.05),两者表达水平随Gyp浓度的增加而升高。四组间p-Smad2、p-Smad3、Smad2、Smad3、TGF蛋白表达水平比较,差异均有统计学意义(均P <0.05)。与空白对照组比较,低浓度Gyp组前述指标表达水平差异均无统计学意义(均P> 0.05),各指标其余两两组间比较差异均有统计学意义(均P <0.05)。结论 Gyp可抑制体外人结肠癌细胞增殖且可促进其凋亡,其可能对TGF-β/Smad信号通路有影响。Objectives To investigate the effect of gypenoside(Gyp) on proliferation and apoptosis of human colon cancer cells and its possible mechanism. Methods Human colon cancer cell SW480 was cultured in vitro and divided into control group,low gyp concentration group(50 μmol/L), medium gyp concentration group(100 μmol/L) and high gyp concentration group(200μmol/L). Cell growth was observed by clonal formation experiment. Cell proliferation was measured by MTT method. Apoptosis was detected by flow cytometry. Expression of proliferation apoptosis protein and protein related to TGF-β/Smad signaling pathway was detected by Western blot. Results There was a significant difference among the four groups in the rate of cell clone formation(P < 0.05), and the rate decreased with the increase of gyp concentration. There were significant differences in the inhibition rate at 24 h and 48 h and apoptosis rate among the four groups(P < 0.05), and the inhibition rate and apoptosis rate increased with the increase of gyp concentration. Levels of expression of Ki67 protein and Bcl-2 protein differed significantly among the four groups(P < 0.05), and levels decreased with the increase of gyp concentration. Levels of expression of Caspase-3 and Bax protein differed significantly among the four groups(P < 0.05), and levels increased with the increase of gyp concentration. There were significant differences in the expression of p-Smad2, p-smad3, Smad2, Smad3 and TGF among the four groups(P < 0.05), pair-wise comparisons showed significant differences expect for comparison between control group and low Gyp concentration group(P > 0.05). Conclusion Gyp could inhibit the proliferation of human colon cancer cells and promote their apoptosis in vitro. Its may have an effect on TGF-β/Smad signaling pathway.
