摘 要:目的探讨miR-155在溃疡性结肠炎(UC)中对外周血Th17细胞的分化及功能的调控作用,阐明miR-155在UC中的作用机制。方法选取2016年6月至2017年6月住院的活动性UC患者16例,免疫磁珠分选出外周血CD4^+T细胞,分为对照组、miR-155组及miR-155抑制组,分别进行培养及干预后,流式细胞仪测定Th17细胞的比例,RT-PCR测定细胞miR-155-3p的表达,ELISA测定培养上清白细胞介素(IL)-17A及IL-6水平。结果各组细胞干预及培养24 h后,Th17细胞的比例[(16.73±4.41)%vs(5.69±1.78)%]及miR-155-3p的表达水平[(1.97±0.56)vs(0.89±0.33)],miR-155组均显著高于对照组(P<0.01),miR-155抑制组Th17细胞的比例、miR-155-3p的表达水平明显低于对照组[(2.03±0.78)%vs(5.69±1.78)%,(0.57±0.16)vs(0.89±0.33),P<0.01]。miR-155组培养上清中IL-17A和IL-6水平也显著高于对照组和miR-155抑制组,差异有统计学意义(P<0.01)。结论miR-155主要通过促进Th17细胞的分化及功能,参与对CD4^+T淋巴细胞的调控,在UC发病机制中发挥着重要的作用。Objective To investigate the role of miR-155 in the differentiation and function of Th17 cells in peripheral blood of patients with ulcerative colitis(UC)to clarify the mechanism of miR-155 in UC.Methods Sixteen patients with active UC from June 2016 to June 2017 were enrolled.By immunomagnetic beads,CD4^+T lymphocytes were isolated from mononuclear cells from the peripheral blood of UC patients and were divided into three groups:control group,miR-155 group and miR-155 inhibition group.The proportion of Th17 cells was detected by flow cytometry,the expression of miR-155-3 p in cells was determined by RT-PCR,and the levels of interleukin(IL)-17 A and IL-6 in supernatant of cultured peripheral CD4^+T lymphocytes were measured using enzyme-linked immunosorbent assay(ELISA).Results After Th17 cells being cultured and intervented for 24 hours,the proportion of Th17 cells[(16.73±4.41)%vs(5.69±1.78)%]and the expression of miR-155(1.97±0.56 vs 0.89±0.33)significantly increased in miR-155 group compared with control group(P<0.01),and the proportion of Th17 cells[(2.03±0.78)%vs(5.69±1.78)%]and the expression level of mir-155-3 p(0.57±0.16 vs 0.89±0.33)in miR-155 inhibition group were significantly lower than those in control group(P<0.01).The levels of IL-17 A and IL-6 in the cultured supernatant in miR-155 group were also significantly higher than those in control group and in miR-155 inhibition group(all P<0.01).Conclusion miR-155 is involved in the regulation of CD4^+T lymphocytes by promoting the differentiation and function of Th17 cells and plays an important role in the pathogenesis of UC.
