摘  要:目的肺癌是中国发病率和死亡率最高的恶性肿瘤,治疗及预后并没有取得太大的进展。研究发现转录因子(transcription factor,TF)与肺癌的发生发展甚至预后关系密切。但在组织水平上大规模鉴定TF存在难度,本文首次利用catTFRE技术对肺癌及癌旁组织的TF进行富集,并初步探讨这些变化调控的生物过程。方法取肺癌及癌旁组织样本,分别提取其的核蛋白,利用TF富集技术catTFRE,对癌及癌旁组织的TF进行富集,同时运用蛋白质组学手段,对表达差异的TF进行动态描绘和定量分析。对于鉴定到的发生变化的TF及蛋白,通过蛋白质组学方法iBAQ、KEGG信号通路等进行生物信息学分析。结果癌症组平均鉴定到1431种蛋白,平均223种TF;癌旁组中平均鉴定到1383种蛋白,平均191种TF。癌症组与癌旁组相比,表达上调的蛋白和TF分别有378种和有108种,表达下调的蛋白和TF分别有245种和50种。对差异表达的TF及蛋白分析发现,如转录因子NFATc1、FOXA1等,VEGF信号通路、Notch信号通路等都与肺癌的发生发展相关。结论catTFRE可高效富集肺癌及癌旁组织的TF,为接下来大样本、高数量鉴定肺癌组织的TF提供了技术支持,为未来对大规模肺癌的深入研究提供一些重要的依据和基础。Objective Lung cancer has the highest morbidity and mortality in China.There has been little progress in treatment and prognosis of lung cancer.It has been found that transcription factor(TF)is closely related to the occurrence,development and even prognosis of lung cancer.However,it is difficult to identify TFs on a large scale at the tissue level.In this paper,catTFRE technology was used to enrich TFs in lung cancer and adjacent tissues for the first time,and the biological processes of these changes were preliminarily explored.Methods Nucleoproteins were extracted from lung cancer and adjacent tissues respectively.TFs was enriched by TF enrichment technology-catTFRE.At the same time,differential expression of TFs were dynamically depicted and quantitatively analyzed by proteomics.Bioinformatics analysis of identified altered TFs and protein by iBAQ and KEGG signaling pathway.Results The average number of TFs was 1431 in the cancer group and 1383 in the para-cancer group,with an average of 191 TFs.There were 378 up-regulated proteins and 108 down-regulated TFs in the cancer group compared with the adjacent cancer group,and 245 down-regulated proteins and 50 down-regulated TFs in the cancer group.The analysis of differentially expressed TFs and protein showed that transcription factors such as NFATc1,FOXA1,and so on,as well as VEGF signaling pathway and Notch signaling pathway were related to the occurrence and development of lung cancer.Conclusion catTFRE can efficiently enrich TF in lung cancer and adjacent tissues.It provides technical support for identifying TF in large samples and large quantities of lung cancer tissues,and provides some important basis for further research on large-scale lung cancer in the future.
