摘 要:目的对SHOX基因进行生物信息学及适应性进化分析,获得氨基酸的基本结构特征和理化性质,研究正选择位点。方法以人SHOXa蛋白序列为模板,应用相关的生物软件进行理化性状、空间结构研究,利用PAML程序检测其SHOXa基因的适应性进化。结果人SHOXa蛋白为292个氨基酸,不具备跨膜结构和信号位点,但有多个磷酸化位点。该蛋白与其他灵长类动物SHOX基因具有较高的同源性,适应性进化分析未发现显著正选择位点,但发现1个潜在的正选择位点(20G)。结论通过对SHOX基因的系统分析,研究其进化特点,为各类矮身材的机制研究提供参考。Objective To analyze the bioinformatics and adaptive evolution of SHOX gene,to obtain basic structural characteristics and physicochemical properties of this protein,and to study the positive selection sites.Methods With the human SHOXa protein sequence as a template,the physical and chemical characteristics,and spatial structure were studied through the biological software.The adaptive evolution of SHOXa gene was detected by PAML program.Results The human SHOXa gene encoded a length of 292 amino acid without transmembrane structure and signal sites,but with multiple phosphorylation sites.Phylogenetic analysis revealed that SHOXa gene of human was similar with primates.No site was found under positive selection in adaptive evolution analysis,but one potential positive selection site(20 G)was detected.Conclusion Based on the systematic analysis and evolutionary characteristics of SHOXa gene,our research provides useful information to explore the mechanism of various short stature phenotypes.
