摘  要:目的分析GSK3β对食管癌KYSE450细胞迁移、侵袭和克隆增殖方面的调控作用。方法采用Wound-healing、Transwell、平板克隆形成实验等方法检测GSK3β对食管癌KYSE450细胞迁移、侵袭和克隆增殖方面调控作用的影响,并分析其调控作用及意义。结果在食管癌KYSE450细胞系中,对GSK3β进行下调时,其迁移、侵袭及克隆增殖方面恶性生物学行为显著减弱,差异有统计学意义(P<0.05)。结论GSK3β高表达能显著促进食管癌KYSE450细胞的恶性生物学行为,抑制GSK3β表达可能成为食管癌预后的评价指标,并为食管鳞癌的治疗提供新的靶点。Objective To analyze the effects of GSK3βon the regulation of migration,invasion and clonal proliferation of esophageal cancer KYSE450 cells.Methods Wound healing,Transwell,plate clone formation experiment and other methods were used to detect the influence of GSK3βon malignant biological behavior of esophageal cancer KYSE450 cells,and to analyze its regulatory role and significance.Results In the KYSE450 cell line of esophageal carcinoma,when the GSK3βwas down-regulated,its malignant biological behaviors in terms of migration,invasion and clone proliferation were significantly reduced,and the difference was statistically significant(P<0.05).Conclusion High expression of GSK3βcan significantly promote the malignant biological behavior of esophageal cancer KYSE450 cells.Inhibition of GSK3βexpression may be a prognostic indicator of esophageal squamous cell carcinoma and provide a new target for the treatment of esophageal squamous cell carcinoma.
