冰醋酸在胸腔液基细胞学制片中的应用研究

摘　　要：目的:探讨冰醋酸作为裂解液在胸腔液基细胞学制片中的应用价值,以期优化液基细胞学制片方法和质量。方法:将疑似肺部病变患者行胸腔液基细胞学检查的标本分为正常组、浑浊液组、血性液组3组,浑浊液组、血性液组分别经50%冰醋酸/生理盐水(1:1),25%冰醋酸/生理盐水(1:3),12.5%冰醋酸/生理盐水(1:7)处理标本后行HE染色,光学显微镜观察染色效果。结果:正常组涂片细胞分部均匀,细胞周围无裂解物,染色清晰;浑浊液组和血性液组各30例标本,分别经冰醋酸50%、25%、12.5%的浓度处理后,涂片分别显示为背景杂乱、清晰、背景异常,差异有显著意义。结论:冰醋酸的浓度影响胸腔液基标本制片效果,以25%冰醋酸处理后的胸腔液基标本制作的涂片质量较好。Objective:To investigate the feasibility and values of glacial acetic acid as auxiliary pyrolysis in the making of pleural liquid-based smears,in order to optimize the liquid based cytology method of its quality.Methods:Liquid-based cytological samples from the patients with suspected pulmonary lesions pleural fluid cytology specimens were divided into three groups:the normal group,blood containing group and turbidity group.The blood-containing group and turbidity group were treated with 50% glacial acetic acid / saline (1:1),25% glacial acetic acid / saline (1:3)and 12.5% glacial acetic acid / saline (1:7) respectively.All the samples from the three groups were rendered into liquid-based smears and stained with HE staining method,and then were observed under light microscope.Results:The cells in the normal group were well distributed over the slides,with no lysate around the cells,and the staining clear.After the concentration of 50%,25% and 12.5% of glacial acetic acid in the turbidity group and the blood fluid group,the smears showed that the background were messy,clear and abnormal,and the difference was significant.Conclusion:The concentrations of acetic acid may affect the smear-making quality,and those treated with 25% glacial acetic acid / saline (1:3) may produce good smear-making quality.
