乳腺癌危险因素和microRNA与雌激素受体表达水平的相关性研究

摘  要:目的:探讨乳腺癌危险因素、microRNA-18a(miRNA-18a)和microRNA-424(miRNA-424)与乳腺癌组织和正常乳腺组织中雌激素受体(ER)表达量的相关性。方法:根据临床经验将乳腺癌可能的危险因素设计成结构式调查问卷,通过调查问卷获取包括文化程度、月经状态等分类变量;用RT-qPCR方法测定25例乳腺癌患者的乳腺癌组织及其对应的正常乳腺组织中miRNA-18a、miRNA-424的相对表达量;利用BCA蛋白浓度测定试剂盒与ELISA试剂盒测定乳腺癌组织和正常乳腺组织中ER的表达量;应用多重线性回归分析miRNA与ER表达的关系。结果:在双侧α=0.05检验水准下,不同劳动类型和直系亲属是否有良性乳腺病史皆对患者ER表达水平有影响(P<0.05)。ER表达水平受ΔmiRNA-18a与ΔmiRNA-424乘积交互项的影响(F=10.837,P=0.005,R^2=0.367)。结论:体力劳动和直系亲属无良性乳腺病史,miRNA-18a和miRNA-424共同作用可能通过影响患者ER的表达水平在乳腺癌发生发展过程中发挥作用;可能可以通过改变劳动方式、根据直系亲属有无良性乳腺病史进行提前预防;miRNA-18a和miRNA-424可能成为治疗乳腺癌的新靶标。Objective: To investigate the relationship between the risk factors of breast cancer, microRNA-18 a(miRNA-18 a) and microRNA-424(miRNA-424) and the expression of estrogen receptor(ER) in breast cancer and normal breast tissues. Methods:According to the clinical experience,the possible risk factors of breast cancer were designed into a structured questionnaire,which was used to obtain the classified variables including education level and menstrual state. The relative expression of miRNA-18 a and miRNA-424 in 25 cases of breast cancer and their corresponding normal breast tissues were measured by RT-qPCR. ER expression in breast cancer and normal breast tissues were determined by BCA protein concentration assay kit and ELISA Kit. The relationship between miRNA and ER expression were analyzed by multiple linear regression. Results:At the level of bilateral α=0.05,the expression of ER was affected by different types of labor and whether the lineal relatives had a history of benign breast(P<0.05). ER expression level was affected by the interaction term of product of ΔmiRNA-18 a and ΔmiRNA-424(F=10.837,P=0.005,R2=0.367). Conclusion:The combined effects of physical labor,no history of benign breast disease in direct relatives,miRNA-18 a and miRNA-424 may play a role in the development of breast cancer by affecting the expression of ER in patients. Breast cancer may be prevented in advance according to whether the immediate family has a history of benign breast. MiRNA-424 and miRNA-18 a may become the new target for breast cancer treatment.
