摘 要:目的探讨碱性离子水联合双歧杆菌三联活菌预防高脂饮食所致大鼠非酒精性脂肪性肝病的作用及机制。方法清洁级SD雄性大鼠60只,随机分为5组,每组12只。空白对照组:普通饲料+普通饮水;模型组:高脂饲料(88%普通饲料+10%猪油+2%胆固醇)+普通饮水;实验A组:高脂饲料+饮碱性离子水,实验B组:高脂饲料+普通饮水+双歧杆菌,实验C组:高脂饲料+饮碱性离子水+双歧杆菌;抽空腹静脉全血检测血清谷丙转氨酶(ALT)、谷草转氨酶(AST)、甘油三酯(TG)、总胆固醇(TC)、血糖(Glu)及尿酸(UA);采用双抗体夹心ELASA法检测空腹胰岛素(FINS)、超氧化物歧化酶(SOD)、丙二醛(MDA)、白细胞介素1β(IL-1β)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α);于造模第12周取肝脏组织行HE染色,确定病理诊断。结果模型组ALT、AST、TG、TC、Glu、UA、FINS、MDA、IL-1β、IL-6、TNF-α较空白对照组显著升高,SOD显著降低(P均<0. 01);实验组A、B、C三组各项指标较模型组均有改善,实验C组各项指标改善均显著优于实验组A及B(P均<0. 01)。结论碱性离子水及双歧杆菌三联活菌均有减轻NAFLD肝脏脂肪变性,提高抗氧化能力,改善肝功能的作用,两者联合疗效可显著增强。Objective To investigate the effect and mechanism of alkaline ionized water combined with bifidobacteria on the prevention of nonalcoholic fatty liver disease in rats. Methods A total of 60 male SD rats were randomly divided into 5 groups, 12 rats in each. The blank control group was fed with regular feed and water. The model group was treated with high fat diet( 88% normal diet + 10% lard + 2% cholesterol) and regular water. The group A was treated with high fat diet and alkaline ionized water. The group B was fed with high fat feed, regular water and bifidobacteria. The group C was treated with high-fat feed, alkaline ionized water and bifidobacteria. After twelve weeks, animals were sacrificed, and liver tissue was processed for HE staining to confirm the pathological diagnosis. Results The levels of ALT, AST, TG, TC, Glu, UA, FINS, MDA, IL-1, IL-6 and TNF-αwere significantly increased in the model group compared with the control group, while SOD was significantly decreased( P < 0. 01). All the indicators in the group A, B and C were improved compared with those in the model group. The improvement of indicator in the group C was better than that in group A and B. Conclusion Both alkaline ionized water and bifidobacteria can alleviate liver steatosis and improve antioxidant capacity and liver function in NAFLD. The combination of the two significantly enhanced the efficacy.
