曲普瑞林抑制子宫平滑肌瘤细胞的性激素受体表达、诱导瘤细胞凋亡

摘　　要：目的研究曲普瑞林抑制子宫肌瘤生长的可能机制。方法利用心肌素过表达刺激人源子宫平滑肌瘤细胞,并用曲普瑞林干预,采用免疫荧光观察SM22α表达,采用Western blot检测SM22α及α-SMA表达水平;建立ELT-3细胞的裸鼠皮下移植瘤模型后,采用腹腔注射曲普瑞林(300 mg/kg/d),每周3次,连续干预4周,检测瘤体质量,并通过免疫组织化学检测雌激素受体α(estrogen receptorα,ERα)、孕激素受体A(progesterone receptor A,PR-A)及孕激素受体B(progesterone receptor B,PR-B)、凋亡相关蛋白Caspase-3及增殖相关蛋白Ki67的表达水平。结果心肌素可明显上调人源平滑肌瘤细胞的SM22α及α-SMA表达水平,曲普瑞林可明显降低由心肌素诱导的SM22α及α-SMA水平增高和裸鼠皮下成瘤组织ERα、PR-A及PR-B表达水平,上调Caspase-3及下调Ki67水平。结论曲普瑞林通过减少性激素受体,上调Caspase-3和下调Ki67抑制肿瘤生长。Objective To study possible mechanism by which triptorelin inhibits the growth of uterine leiomyoma.Methods Human uterine leiomyoma cells were stimulated by overexpression of myocardin and then were treated with triptorelin in this process.The expression of SM22αwas observed by immunofluorescence,and the expression levels of SM22αandα-SMA were detected by western blot.Subcutaneous xenograft model of ELT-3 cells in nude mice was established,then these mice were intraperitoneally injected with triptorelin(300 mg/kg/d)3 times per week for 4 consecutive weeks.Tumor weight were detected.The expression levels of estrogen receptorα(ERα),progesterone receptor A(PR-A),progesterone receptor B(PR-B),apoptosis-related protein Caspase-3 and proliferation-related protein Ki67 were detected by immunohistochemistry.Results Myocardin significantly up-regulated the expression levels of SM22αandα-SMA in human uterine leiomyoma cells,while triptorelin significantly reduced the increase of SM22αandα-SMA levels induced by cardiomycin and the expression levels of ERα,PR-A and PR-B in subcutaneous tumorigenic tissues of nude mice,as well as up-regulated the levels of Caspase-3 and down-regulated the levels of Ki67.Conclusion Triptorelin inhibits tumor growth by decreasing hormone receptors,up-regulating caspase-3 and down-regulating Ki67.
