摘 要:目的观察Wnt1、β-catenin、H3K9ac在膀胱肿瘤组织与正常癌旁组织的表达,并分析组蛋白乙酰化表达量的高低与Wnt、β-catenin表达的相关性,探讨Wnt信号通路异常对组蛋白乙酰化表达水平的影响。方法收集来自本院膀胱肿瘤全切标本,分别取肿瘤组织与正常癌旁组织各32例,然后通过Western blot检测Wnt1、β-catenin、H3K9ac相对蛋白量的表达;RT-qPCR检测Wnt1、β-catenin、H3K9ac mRNA的表达量。统计学分析肿瘤组织与正常癌旁组织之间的差异;组蛋白乙酰化与Wnt、β-catenin表达量的相关性。结果Western blot结果显示,与正常癌旁组织相比,Wnt1、β-catenin在膀胱肿瘤组织中高表达,差异有统计学意义(P<0.05);H3K9ac在膀胱肿瘤组织中低表达,差异有统计学意义(P<0.05);组蛋白乙酰化与Wnt、β-catenin表达量呈正相关,差异无统计学意义。结论膀胱癌肿瘤组织中Wnt1、β-catenin呈高表达,H3K9ac呈低表达;组蛋白乙酰化与Wnt、β-catenin的表达量无相关性,组蛋白乙酰化修饰水平降低可能与膀胱癌的发生发展相关。Objective To observe the expression of Wnt1,β-catenin and H3K9ac in bladder tumor tissues and normal adjacent tissues,to analysis the correlation between the expression level of histone acetylation,Wnt andβ-catenin to investigate the effect of abnormal Wnt signaling pathway on the expression level of histone acetylation.Methods The total specimens of bladder tumors from our hospital were collected,and 32 cases of tumor tissue and normal adjacent tissues were taken,then the expression of Wnt1,β-catenin and H3K9ac relative protein were detected by Western blot,and the expression levels of Wnt1,β-catenin and H3K9ac mRNA were detected by RT-qPCR.Statistical analysis of the difference between tumor tissue and normal adjacent tissue,the correlation between histone acetylation and the expression levels of Wnt andβ-catenin.Results Western blot results showed that Wnt1 andβ-catenin were highly expressed in bladder tumor tissues compared with normal adjacent tissues(P<0.05).H3K9ac was lowly expressed in bladder tumor tissues and the difference was statistically significant(P<0.05).Histone acetylation was positive correlated with the expression of Wnt andβ-catenin,and there was no statistically significant difference.Conclusion Wnt1 andβ-catenin were highly expressed in bladder cancer tissues,and H3K9ac was down-regulated.Histone acetylation was not associated with the expression of Wnt andβ-catenin,and the decrease of histone acetylation level may be related to the occurrence and development of bladder cancer.
