摘 要:目的:探讨广西红水河流域长寿群体血清β-珠蛋白基因(HBB)mRNA表达水平与血糖、血脂及肾功能代谢指标的关系,以了解其在机体衰老中的作用。方法:选取广西红水河流域居民303例,分为长寿组(年龄90~102岁,n=107)、老年组(年龄65~84岁,n=68)和青壮年组(年龄20~64岁,n=128)。检测胆固醇(TC)、甘油三酯(TG)、肌酐(Cr)、肾功能等指标,实时荧光定量PCR(RT-qPCR)法测定各组血清HBB mRNA的表达情况,Spearman相关分析法分析HBB mRNA表达水平与血糖、血脂及肾功能代谢指标的关系。结果:与老年组比较,长寿组血清同型半胱氨酸(Hcy)、免疫球蛋白G(IgG)、尿素氮(BUN)、Cr水平明显升高,空腹血糖(FBG)、TC、高密度脂蛋白(HDL)、载脂蛋白A(ApoA)、载脂蛋白B(ApoB)水平降低;与青壮年组比较,长寿组血清TG、TC、HDL、ApoA、ApoB水平明显降低(P<0.05);长寿组血清HBB mRNA相对表达量与青壮年组和老年组相比均显著降低(P<0.05);血清HBB mRNA表达水平与ApoA、FBG呈正相关关系,与IgG、Hcy、Cr、BUN呈负相关关系。结论:血清HBB mRNA可能与代谢性指标相互作用进而影响长寿群体衰老进程。Objective:To investigate the effect of serumβ-globin(HBB)mRNA expression level on blood glucose and blood lipid and renal metabolism index in longevity population in Guangxi Hongshuihe region.Methods:303 residents in Guangxi Hongshuihe region were selected as the study objects,and divided into longevity group(the age ranges from 90 to 102,n=107),elderly group(the age ranges from 65 to 84,n=68)and young group(the age ranges from 20 to 64,n=128).Clinical parameters such as cholesterol(TC),triglyceride(TG),creatinine(Cr)and renal function were detected.The expression of serum HBB mRNA in each group was measured by real-time quantitative PCR(RT-qPCR).Spearman correlation analysis was used to analyze the relationship between HBB mRNA expression level and blood glucose,blood lipid and renal metabolism indexes.Results:Compared with the elderly group,the serum levels of homocysteine(Hcy),immunoglobulin G(IgG),urea nitrogen(BUN)and Cr in the longevity group were increased significantly,while the levels of fasting blood glucose(FBG),TC,high-density lipoprotein(HDL),apolipoprotein A(ApoA)and apolipoprotein B(ApoB)were decreased.Compared with the young group,the serum levels of TG,TC,HDL,ApoA and ApoB in the longevity group were significantly decreased(P<0.05).The expression of HBB mRNA in the longevity group was lower than young and old groups(P<0.05).The expression level of serum HBB was positively correlated with ApoA and FBG,and negatively correlated with IgG,Hcy,Cr and BUN.Conclusion:HBB mRNA may interact with metabolic risks and further influence the aging process of long-lived individuals.
