摘 要:针对一般生物表面活性剂产生菌产量低、生产成本高的问题,研究了FeSO4和MnSO4对苍白杆菌(Ochrobactrum sp.)发酵产生物表面活性剂的影响,考察在30℃、160 r/min摇床发酵过程中细胞浓度、pH值、糖浓度以及谷氨酰胺合成酶和谷草转氨酶活性等重要参数的变化。结果表明,在蔗糖10 g/L、磷酸氢钾3.4 g/L、磷酸氢钠1.5 g/L、硝酸钠4.0 g/L和酵母浸粉0.2 g/L的培养基中,添加0.4 mmol/L FeSO4可提高谷草转氨酶活性并加快生物表面活性剂积累,产量(1.75 mg/L)提高了7.5倍;添加0.3 mmol/L MnSO4可提高谷氨酰胺合成酶活性,使生物表面活性剂产量(2.21 mg/L)提高8.6倍。To solve the problems of low yield and high production cost for general biosurfactant-producing bacteria,the effects of FeSO4 and MnSO4 on biosurfactant production by Ochrobactrum sp.fermentation were investigated.The changes of main parameters such as cell concentration,pH,sugar concentration,glutamine synthetase and glutamic-oxalacetic transaminease were investigated during the fermentation at 30℃and 160 r/min.The results showed that in the medium with sucrose 10 g/L,potassium hydrogen phosphate 3.4 g/L,sodium hydrogen phosphate 1.5 g/L,sodium nitrate 4.0 g/L and yeast extract 0.2 g/L,the addition of FeSO40.4 mmol/L increased the activities of glutamic-oxalacetic transaminease and the biosurfactant yield(1.75 mg/L)increased by 7.5 times;The addition of MnSO40.3 mmol/L increased glutamine synthetase activity and the biosurfactant yield(2.21 mg/L)increased by 8.6 times.
