基于Cancer Browser数据库分析促癌基因HMGA2在肾癌中的表达及其临床意义

摘　　要：目的探究HMGA2基因在肾癌中的表达及其对上皮间质转化的影响。方法通过Cancer Browser数据库检测HMGA2的表达与肾癌TNM分期、Staging分期的相关性,并明确HMGA2与Zeb1、Zeb2在肾癌中的相关性;随后通过Western blot实验和实时定量PCR检测HMGA2在肾癌组织、癌旁正常组织、肾癌细胞系以及正常肾小管上皮细胞内的蛋白及mRNA表达水平;通过Cancer Browser数据库检测HMGA2与Zeb1、Zeb2在肾癌中表达的相关性,并通过Western blot实验检测肾癌组织与癌旁正常组织、肾癌细胞786-O与正常肾小管上皮细胞HK2中Twist1、Twist2、Zeb1、Zeb2、HMGA2的蛋白表达水平;最后通过Western blot实验和实时定量PCR检测敲低HMGA2后肾癌细胞786-O内Twist1和Twist2的表达水平。结果随着肾癌的TNM分期以及Stage分期的逐渐增高,HMGA2的表达亦显著增高,且差异有统计学意义(P<0.01)。而HMGA2在肾癌组织及细胞内的表达显著高于癌旁正常组织及正常肾小管上皮细胞(P<0.001);不管在肾癌组织还是在肾癌细胞内,HMGA2与Twist1、Twist2均呈高表达,而Zeb1和Zeb2的表达无明显变化(P=0.3137,0.5820)。而Western blot和实时定量PCR结果显示敲低HMGA2可下调肾癌细胞内Twist1和Twist2的蛋白和mRNA表达水平。结论HMGA2与肾癌的TNM分期和Stage分期呈正相关。而HMGA2在肾癌组织及细胞内呈高表达,且HMGA2可正向调控Twist1和Twist2的表达。Objective To explore the expression of HMGA2 in renal cell carcinoma and the effect of HMGA2 on epithelial-mesenchymal transition.Methods Cancer Browser database was used to detect the correlation between the expression of HMGA2 and the TNM,staging classification of renal cell carcinoma,and demonstrate the correlation between the HMGA2 and Zeb1,Zeb2 in renal cell carcinoma.Subsequently,Western blot and real-time PCR were used to detect the protein and mRNA levels of HMGA2 in renal cell carcinoma tissue,cell lines,normal tissue and renal tubular epithelial cell.Then Western blot was used to detect the protein levels of Twist1,Twist2,Zeb1,Zeb2 and HMGA2 in renal cell carcinoma tissue,adjacent normal tissue,786-O cell line,and renal tubular epithelial HK2 cell line.Finally,Western blot and real-time PCR were performed to detect the expression of Twist1 and Twist2 in 786-O cell line after HMGA2 knockdown.Results The HMGA2 expression was significantly increased as the TNM,staging classification of renal cell carcinoma increased gradually(P<0.01).The expression of HMGA2 was higher in the renal cell carcinoma tissues and cells than that in normal paracancerous tissues and normal renal tubular epithelial cell(P<0.001).And HMGA2 and Twist1,Twist2 were highly expressed in the renal cell carcinoma tissues and cells,but Zeb1 and Zeb2 showed no obvious change(P=0.3137,0.5820).Conclusion HMGA2 has a positive correlation with TNM,staging classification of renal cell carcinoma.And HMGA2 is highly expressed in the renal cell carcinoma.In addition,HMGA2 could positively regulate the expression of Twist1 and Twist2.
