摘  要:目的观察口服黑升麻提取物对围绝经期雌鼠糖脂代谢的调节作用。方法选取30只处于围绝经期的雌鼠随机分为3组,分别记为黑升麻组、雌激素组和对照组,另选取6月龄SD雌鼠10只,记为青年对照组。黑升麻组雌鼠每日口服黑升麻提取物60 mg/kg,雌激素组雌鼠每日口服雌激素0.8 mg/kg,对照组和青年对照组雌鼠每日口服5%生理盐水10 mL/kg。各组雌鼠在入组时(T0)及治疗2个月后(T1),称量大鼠体质量;采集鼠尾静脉血,检测糖脂代谢指标:应用全自动生化分析仪检测血清空腹血糖(FBG)、总胆固醇(TC)、甘油三酯(TG),采用ELISA法检测血清空腹胰岛素(FINS)、8点皮质醇、超敏C反应蛋白(hs-CRP),计算胰岛素敏感性指数(ISI,ISI=1/[Log(FINS)+Log(FBG)])及胰岛素抵抗指数(HOMA-IR,HOMA-IR=FBG×FINS/22.5)。结果与青年对照组相比,对照组雌鼠在T1时体质量、FBG、FINS、HOMA-IR、TC、TG、8点皮质醇、hs-CRP均升高(P均<0.05),ISI降低(P<0.05)。与对照组相比,黑升麻组雌鼠在T1时体质量、FBG、FINS、HOMA-IR、TG、8点皮质醇、hs-CRP均降低(P均<0.05),ISI升高(P<0.05);雌激素组雌鼠在T1时体质量、FBG、TG、hs-CRP均降低(P均<0.05)。与雌激素组相比,黑升麻组雌鼠在T1时FINS、HOMA-IR、8点皮质醇均降低(P均<0.05),ISI升高(P<0.05)。结论围绝经期雌鼠糖脂代谢指标紊乱;黑升麻提取物具有调节围绝经期雌鼠糖脂代谢指标的作用;与雌激素相比,黑升麻提取物可有效缓解围绝经期雌鼠的8点皮质醇水平及HOMA-IR。Objective To observe the effect of black cohosh extract on glucose and lipid metabolism in perimenopaus-al female rats.Methods Thirty perimenopausal female rats were randomly divided into three groups:black cohosh group,estrogen group,and control group.Another ten 6-month-old SD female rats were selected and recorded as the young control group.The female rats in the black cohosh group took 60 mg/kg black cohosh extract daily,the female rats in the estrogen group took 0.8 mg,/kg estrogen daily,and the female rats in the control group and the young control group took 5%normal saline 10 mL/kg daily.At the time of entering groups(T0)and 2 months after treatment(T1),the body weight of female rats in each group was weighed;the tail vein blood of rats was collected and the metabolic indexes of glucose and lipid were detected:FBG,TC and TG were detected by automatic biochemical analyzer;FINS,8-point cortisol and hs-CRP were detected by ELISA;and we calculated insulin sensitivity index(ISI,ISI=1/[Log(FINS)+Log(FBG)])and insu lin resistance index(HOMA-IR,HOMA-IR=FBG x FINS/22.5).Results Compared with the young control group,the body weight,FBG,FINS,HOMA-IR,TC,TG,8-point cortisol,hs-CRP of female rats in the control group increased more(P<0.05),ISI decreased more(P<0.05).Compared with the control group,the body weight,FBG,FINS,HOMA-IR,TG,8-point cortisol and hs-CRP of the female rats in the black cohosh group decreased(all P<0.05),ISI increased(P<0.05);the body weight,FBG,TG and hs-CRP of the female rats in the estrogen group decreased(all P<0.05).Compared with the estrogen group,FINS,HOMA-IR and 8-point cortisol in the black cohosh group decreased(all P<0.05),and ISI increased(P<0.05).Conclusions Perimenopausal female rats have disordered glucose and lipid metabolism indicators;black cohosh extract can regulate the glycolipid metabolism of perimenopausal female rats;compared with estrogen,black cohosh extract can effectively alleviate 8-point cortisol level and HOMA-IR of perimenopausal female rats.
