摘  要:目的:建立川芎配伍水煎液HPLC特征图谱及有效成分阿魏酸和藁本内酯的含量测定方法,为阐明川芎临床配伍用药提供物质基础依据。方法:制备天麻、细辛、丹参、连翘、赤芍、党参单煎液及与川芎配伍的合煎液,采用HPLC-DAD建立川芎及川芎配伍水煎液HPLC特征图谱并检测各组水煎液中阿魏酸、藁本内酯的含量,以单煎液为对照,统计分析诸药与川芎配伍后有效成分的含量变化。结果:获得川芎及川芎药对HPLC特征图谱,通过对共有特征峰的归属,发现药对特征图谱与单药特征图谱比较,基本为两单味药特征峰的加和,没有色谱峰的增减;川芎单煎组阿魏酸与藁本内酯含量分别为1.065 mg·g-1和0.062 mg·g-1,细辛、丹参、赤芍、党参与川芎药对配伍阿魏酸的含量为1.148~1.376 mg·g-1,较川芎单煎组均显著升高,天麻与川芎药对配伍阿魏酸的含量为0.248 mg·g-1,显著降低,而连翘配伍组无显著差异;药对配伍各组藁本内酯含量为0.077~0.616 mg·g-1,较川芎单煎组均显著升高。结论:天麻、细辛、丹参、连翘、赤芍、党参与川芎药对配伍,均对川芎中有效成分的煎出有影响,而未发现有新药效成分的增减,初步探明川芎药对配伍规律,为川芎的进一步深入研究及临床应用提供依据和参考。Objective:To establish the HPLC method to determine the characteristic spectrum of Chuanxiong compatible decoction and the content of its active components including ferulic acid,ligustilide in order to provide a material basis for clarifying the compatibility of Chuanxiong in clinic.Methods:decoction of Tianma,Xixin,Danshen,Lianqiao,Chishao,Dangshen and their admixture decoction with Chuanxiong were prepared for HPLC-DAD to acquire the characteristic spectrum of Chuanxiong decoction and its admixture decoctions.The contents of ferulic acid and ligustilide in the above decoctions were detected respectively.And the contents of ferulic acid and ligustilide in admixture decoctions were compared with those in Chuanxiong decoction.Results:Compared with the characteristic spectrum of Chuanxiong decoction,the characteristic peak of every admixture decoction was basically the sum of the two characteristic peaks of the two single drugs and there was no increase or decrease of chromatographic peaks.The contents of ferulic acid and ligustilide in Chuanxiong decoction were 1.065 mg·g-1 and 0.062 mg·g-1,respectively.The content of ferulic acid in Xixin-Chuanxiong decoction,Danshen-Chuanxiong decoction,Chishao-Chuanxiong decoction,DangshenChuanxiong decoction was in the range of 1.148 to 1.376 mg·g-1,which was significantly higher than that of Chuanxiong decoction.The content of ferulic acid in Tianma-Chuanxiong decoction(0.248 mg·g-1)was significantly lower than that of Chuanxiong decoction.There was no significant difference between the content of ferulic acid in Lianqiao-Chuanxiong decoction and Chuanxiong decoction.The content of ligustilide in the admixture decoction was in the range of 0.077~0.616 mg·g-1,which was significantly higher than that of Chuanxiong decoction.Conclusion:It was indicated that the active components in Chuanxiong decoction changed when Chuanxiong was used in combination with Tianma,Xixin,Danshen,Lianqiao,Chishao,Dangshen respectively,but there was no new active components found in admixture de
