摘 要:目的探讨可降解聚酰胺树枝状大分子(SS-PAMAM)复合基因载体对于人肝癌细胞系HepG2细胞蛋白表达及细胞增殖迁移的影响。方法将人肝癌细胞学HepG2细胞分为观察组和对照组,观察组给予复合型PAMAM基因载体处理,对照组给予空载体处理,蛋白质印迹法检测CDH13蛋白表达,采用四甲基偶氮唑盐(MTT)法和划痕实验检测肿瘤细胞的增殖和迁移能力。结果观察组HepG2细胞CDH13蛋白表达为(0.981±0.019),明显高于对照组的(0.420±0.017),差异有统计学意义(P<0.05);观察组HepG2细胞24 h增殖吸光度值及划痕愈合率分别为(0.554±0.013)和(0.198±0.011),均明显低于对照组(P<0.05)。结论复合型PAMAM基因载体可使人肝癌HepG2细胞的增殖和迁移能力减弱。Objective To investigate the effect of complex PAMAM gene vector on protein expression and cell proliferation of human hepatoma cell line HepG2.Methods HepG2 cells were divided into observation group and control group.The observation group was treated with the composite PAMAM gene vector,the control group was treated with empty vector,and the proliferation and migration ability of tumor cells were detected by MTT assay and scratch test.Results The expression of CDH13 protein of HepG2 cells in the observation group was(0.981±0.019),which was significantly higher than that in the control group(0.420±0.017).The difference was statistically significant(P<0.05).The A value and scratch healing rate of HepG2 cells in the observation group were(0.554±0.013)and(0.198±0.011),respectively,which were significantly lower than those in the control group(P<0.05).Conclusion The composite PAMAM gene vector can attenuate the cell proliferation and migration of human hepatoma HepG2 cells.
