驱蚊香草离体扩繁系统的建立

摘  要:驱蚊香草是一种能够驱赶蚊虫且对人体无害,还具有抗癌、治疗风湿等药理作用的植物,具有良好的应用前景。但是其种子结实率低,而扦插栽培技术对扦插条件要求又高,为了提高驱蚊香草的繁殖效率,以驱蚊香草叶柄为试验材料,选择MS+NAA(0.1、0.2、0.3、0.4、0.5 mg/L)+6-BA(0.5、1.0 mg/L)+蔗糖(20、30 g/L)培养基作为愈伤组织和不定芽的诱导培养基,在不含激素的MS0生根培养基(1/2MS 26.42 g/L+蔗糖30 g/L+琼脂5 g/L)上,利用植物组织培养方法进行了外植体扩繁。结果表明:驱蚊香草最适愈伤组织和不定芽诱导培养基为MS+NAA 0.1 mg/L+6-BA 1.0 mg/L+蔗糖30 g/L,愈伤组织诱导率高达99%,不定芽诱导率达96%;在1个实验周期内,一段1 cm长的叶柄平均可以得到5.97个植株,且再生芽生长健壮。本研究结果可以驱蚊香草快速离体扩繁体系的建立提供技术支持。Pelargonium graveolens can drive away mosquitoes and is harmless to human body.It also has anti-cancer,anti-tumor and other pharmacological effects,and has good application prospect.However,the seed setting rate is low,and the cutting cultivation technique has high requirements for cutting conditions.In order to improve the reproductive efficiency of P.graveolens,the petiole was used as the test material,and MS+NAA(0.1,0.2,0.3,0.4 and 0.5 mg/L)+6-BA(0.5 and 1.0 mg/L)+sucrose(20 and 30 g/L)was selected as an induction medium for callus and adventitious buds.In hormone-free MS0 rooting medium(1/2 MS),explants were propagated by plant tissue culture method using 1/2 MS 26.42 g/L+sucrose 30 g/L+agar 5 g/L.The results showed that the optimum callus and adventitious bud induction medium was MS+NAA 0.1 mg/L+6-BA 1.0 mg/L+sucrose 30 g/L,the callus induction rate was 99%,and the adventitious bud induction rate was 96%.In a experimental cycle,an average of 1 cm long petiole yielded 5.97 plants,and the regenerated shoots grew robustly.The results of this study can provide technical support for the establishment of a rapid in vitro propagation system for P.graveolens.
