摘 要:目的观察蒙药那如三味丸对脊髓损伤模型大鼠细胞凋亡的影响。方法将12只大鼠按随机数字表法分为假手术组、模型组、那如三味片组,每组4只。模型组、那如三味丸组采用改良Allen's法建立脊髓损伤大鼠模型。损伤后72 h处死大鼠,分离提取各组大鼠脊髓细胞,那如三味丸组细胞每孔加入那如三味丸溶液0.3 g/ml,干预24 h后,采用PCR、Western blot检测各组脊髓细胞caspase-3基因及蛋白相对表达,采用流式细胞术分析细胞凋亡情况。结果与模型组比较,那如三味丸组脊髓细胞caspase-3 mRNA[(2.177±0.084)比(3.287±0.098)]、caspase-3蛋白[(1.063±0.157)比(2.180±0.147)]表达显著下调(P<0.05),细胞凋亡率[(11.400±0.953)%比(14.260±1.105)%]下降(P<0.05)。结论那如三味丸可有效降低脊髓损伤大鼠的细胞凋亡率,其作用机制可能与抑制caspase-3蛋白及基因表达有关。Objective To observe the effect of the Mongolian medicine on the apoptosis after spinal cord injury.Methods A total of 12 rats were divided into the sham group,the model group,and the like-3 treatment groups(Naru-3 group),4 in each group.A rat model of spinal cord injury(SCI)was established by modified Allen's method.After 72 h,we sacrificed the rats,separated spinal cord cells,which were filled with 5×10^5 shop in every hole,training for subsequent testing after 24 h.The relative expression of caspase-3 was detected by RT-QPCR and western blot,and apoptosis was analyzed by flow cytometry.Results Compared to the model group,the expression of caspase-3 mRNA(2.177±0.084 vs.3.287±0.098)and caspase-3 protine(1.063±0.157 vs.2.180±0.147)in the Naru-3 group significantly decreased(P<0.05).The apoptosis rate(11.400%±0.953%vs.14.260%±1.105%)of the spinal cord in the Naru-3 group significantly decreased(P<0.05).Conclusions The Naru-3 solution can reduce the rate of spinal cord cell apoptosis after spinal cord injury,its mechanism may be related to the suppressed caspase-3 protein expression.
