摘  要:目的:探讨D-丝氨酸对地卓西平马来酸盐(MK-801)诱导的精神分裂症模型大鼠行为和认知功能的影响。方法:取50只青春期雄性SD大鼠随机均分为5组:MK-801模型(MK)组、MK-801模型+D-丝氨酸低剂量(MK+D1)组、MK-801模型+D-丝氨酸高剂量(MK+D2)组、MK-801模型+利培酮(MK+R)组和生理盐水(NS)组。NS组与其余4组分别给予生理盐水与MK-80114 d造模;造模结束24 h后,NS组与MK组给予生理盐水10 d,MK+D1组、MK+D2组与MK+R组分别接受低剂量与高剂量D-丝氨酸和利培酮干预10 d。在药物干预前后对各组大鼠进行旷场实验、强迫游泳及Y迷宫等行为学实验。结果:与NS组相比,MK-801造模大鼠的旷场实验自主活动及探究行为均增加,强迫游泳实验不动时间延长,Y迷宫实验交替百分率降低(P<0.05或P<0.01)。与MK组相比,药物干预后MK+D1组、MK+D2组与MK+R组的自主活动及探究行为减少,强迫游泳不动时间缩短,Y迷宫实验交替百分率升高(P<0.05或P<0.01)。结论:D-丝氨酸对精神分裂症模型大鼠的阳性症状、阴性症状以及认知损伤中的工作记忆均有一定的改善作用。Objective:To observe the effects of D-serine on the behavioral and cognitive functions of schizophrenic model rats induced by dizocilpine maleate(MK-801). Method:50 male SD rats were randomly assigned into five groups:MK group,MK+D1 group,MK+D2 group,MK+R group and NS group.NS group was exposed to saline,other groups were all exposed to MK-801 for 14 days.After 24 hours,NS group and MK group received normal saline,and the other three groups were treated with low and high D-serine or risperidone for 10 d.All the rats were tested by the open field test,forced swimming test and Y-maze behavior test before and after the drug intervention. Results:Compared with the NS group,locomotor activity and explorative behavior in open-field test and immobility time in the forced swimming test increased,and alternate percentage in Y-maze spontaneous alternation test decreased in the MK-801 model rats(P<0.05 or P<0.01).After medicine intervention,MK+D1,MK+D2 and MK+R groups showed significantly decreased locomotor activity and explorative behavior and immobility time,and increased alternate percentage(P<0.05 or P<0.01),which were different from the MK group. Conclusion:D-serine may have an positive effect on the positive symptoms,negative symptoms and the working memory in cognitive impairment of model rats with schizophrenia.
