冻干富血小板血浆促进小鼠皮肤创面修复的实验研究

摘　　要：目的:对比冻干富血小板血浆(PRP)与新鲜PRP对小鼠皮肤缺损修复的影响。方法:获取新鲜PRP以及制备冻干PRP。用酶联免疫吸附法(ELISA)检测新鲜PRP与冻干PRP释放血管内皮生长因子(VEGF)及碱性成纤维细胞生长因子(bFGF)浓度。将冻干PRP和新鲜PRP作用于小鼠皮肤缺损模型,观察2组皮肤愈合能力。免疫荧光检测两组创面中M2/M1型巨噬细胞比例。Masson染色观察创面胶原新生结果。结果:冻干PRP与新鲜PRP体外生子因子释放速率类似。新鲜PRP早期能够较快促进创面愈合,然而长期观察无明显差异。冻干PRP与新鲜PRP均能有效地促进创面中胶原合成,且能上调创面中M2型巨噬细胞比例。结论:冻干PRP拥有与新鲜PRP类似的生长因子释放结果,且能够有效促进创面愈合,为将来PRP的保存及应用提供了一定的理论支持。Objective:to compare the effects of lyophilized platelet rich plasma(PRP)and fresh PRP on the repair of skin defects in mice.Methods:The concentrations of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF)released from loaded platelet rich plasma microspheres were measured by enzyme-linked immunosorbent assay(ELISA).Lyophilized PRP and fresh PRP were used to observe the skin healing ability of the two groups.The proportion of M2/M1 macrophages in the two groups was detected by immunofluorescence.Masson staining was used to observe the ability of collagen formation.Results:the release rate of PRP was similar to that of PRP in vitro.Fresh PRP can promote wound healing in early stage,but there is no significant difference in long-term observation.Both lyophilized PRP and fresh PRP can effectively promote collagen synthesis in wounds,and can up-regulate the proportion of M2 macrophages in wounds.Conclusion:lyophilized PRP has similar growth factor release results compared with fresh PRP,and can effectively promote wound healing,indicating that lyophilized PRP could be a promising product in the future.
