摘  要:目的探讨三氧化二砷(As2O3)对小鼠B16细胞的生长抑制作用,对p53、Bax、Bcl-2蛋白表达的影响,为临床治疗恶性黑素瘤提供理论和实验依据。方法将不同剂量的As2O3作用于小鼠B16细胞后,吉姆萨染色和荧光染色观察B16细胞的凋亡变化;Western blot检测p53、Bax、Bcl-2蛋白的表达。结果染色后镜下可见As2O3可致B16细胞出现凋亡现象;As2O3能诱导p53、Bax蛋白的表达,抑制Bcl-2蛋白的表达(P<0.01)。结论As2O3能诱导B16细胞凋亡,这一现象可能是通过诱导B16细胞的p53、Bax蛋白表达上调,Bcl-2蛋白表达下降来实现。Objective To investigate the growth inhibition role of arsenic trioxide(As2O3)on mice malignant melanoma B16 cell,and observe the expression of p53,Bax,Bcl-2 protein in B16 cell after induced by As2O3.To provide theory and experiment base for treatment of malignant melanoma.Methods The morphologic changes of B16 cells were detected by staining.The expression of p53,Bax,Bcl-2 protein was detected by Western blot.Results As2O3 caused morphologic change of B16 cells by staining.The expression of Bax and p53 protein was up-regulated in B16 cells by co-culture with As2O3(P<0.01).The expression of Bcl-2 protein was inhibited by As2O3(P<0.01).Conclusion As2O3 can cause morphologic change of B16 cells and promote apoptosis happening,which may be achieved by inducing the up-regulation of p53 and Bax protein expression,the down-regulation of Bcl-2 protein expression in B16 cells.
