摘 要:近年来,我国集约化牛场卵巢静止性乏情率较高,但临床奶牛卵巢静止的发生尚未有完整的预警体系,为临床预测奶牛卵巢静止提供预警指标,选取产后60~90d卵巢静止组和对照组奶牛血液样本,在蛋白组学前期研究的基础上筛选出检测试验奶牛血液中与卵巢静止相关指标。3种能量指标:谷草转氨酶(AST)、葡糖糖(Glu)、非游离性脂肪酸(NEFA);4种生殖激素:雌二醇(E2)、孕酮(P4)、促卵泡生长激素(FSH)、促黄体素(LH);4种关键差异表达蛋白:胰岛素样生长因子结合蛋白(IGFBP-2)、α-2-HS糖蛋白(AHSG)、载脂蛋白A4(ApoA4)、视黄醇结合蛋白4(RBP-4);6种关键酶活性:果糖二磷酸醛缩酶(ALDOB)、L-乳酸脱氢酶(LDHB)、inter-α-胰蛋白酶抑制剂重链H3(ITIH3)、谷胱甘肽过氧化物酶(GPX3)、血清衍生的透明质酸相关蛋白(SPAM1)、丙酮酸激酶(PKM2)。结果显示,经过相关性分析、二元逻辑回归建模以及ROC分析,建立了ApoA4和ITIH3单一指标的预警技术,其预警值分别为ApoA4>28.825μg/L,ITIH3>195.07ng/L。建立了基于ApoA4+ITIH3和ApoA4+ITIH3+E2潜在生物标志物的多指标预警体系,前者预警值为ApoA4>19.55μg/L,ITIH3>191.14ng/L;后者预警值为ApoA4>47.56μg/L,ITIH4>187.80ng/L,E2<69.63ng/L。In recent years,the rate of ovarian quiescence is high in intensive cattle farms in China,and there is no complete warning system for the occurrence of ovarian quiescence in clinical cows.This experiment aims to clinically predict the ovary rest of cows to provide early warning indicators.This experiment collected blood samples of dairy cows from 60to 90days postpartum in the ovary static group and control group.Three energy indexes were selected on the basis of proteomics:aspartate aminotransferase(AST),glucosamine(Glu),and non-dissociative fatty acids(NEFA);four reproductive hormones:estradiol(E2),progesterone(P4),follicle growth hormone(FSH),luteinizing hormone(LH);four key differentially expressed proteins:insulin-like growth factor combination protein 2(IGFBP-2),α-2-HS glycoprotein(AHSG),apolipoprotein A4(ApoA4),retinol binding protein 4(RBP-4);and six key enzyme activity:fructose diphosphate aldolase(ALDOB),L-lactate dehydrogenase(LDHB),inter-α-trypsin inhibitor heavy chain H3(ITIH3),glutathione peroxidase(GPX3),serum-derived hyaluronan-related protein(SPAM1),pyruvate kinase(PKM2).The results showed that after correlation analysis,binary logistic regression modeling and ROC analysis,a single indicator early warning technique for ApoA4and ITIH3 was established.The early warning values were ApoA4>28.825μg/L and ITIH3>195.07ng/L.A multi-index early warning system based on potential biomarkers of ApoA4+ITIH3and ApoA4+ITIH3+E2was established.The former had an early warning value:ApoA4>19.55μg/L,ITIH3>191.14ng/L;the latter has an early warning value:ApoA4>47.56μg/L,ITIH4>187.80ng/L,E2<69.63ng/L.
