摘 要:目的观察复方当归川葛颗粒对偏头痛大鼠血浆NO、NOS、CGRP、ET及脑组织5-HT及DA的影响。方法60只雄性Wistar大鼠随机分为空白对照组、模型对照组、当归川葛颗粒低剂量组(0.9g/kg)、复方当归川葛颗粒中剂量组(1.8g/kg)、复方当归川葛颗粒高剂量组(3.6g/kg)及琥珀酸舒马普坦组(0.05g/kg),每组10只,各组均连续给药7 d。末次给药30 min后,除空白对照组外,其余各组大鼠臀部皮下注射10 mg/kg硝酸甘油,空白对照组大鼠臀部皮下注射等体积生理盐水。造模180min后,各组大鼠腹腔麻醉,分离血浆,测定大鼠血浆NO、NOS、CGRP及ET质量浓度;剖取脑组织,制备组织匀浆液,测定大鼠脑组织5-HT及DA质量浓度。结果中、高剂量复方当归川葛颗粒可以降低偏头痛大鼠血浆NO、NOS、CGRP及ET质量浓度,升高大鼠脑组织中5-HT及DA质量浓度。结论复方当归川葛颗粒对硝酸甘油诱导的大鼠偏头痛具有预防性治疗作用,其作用机制可能与调节大鼠血浆NO、NOS、CGRP及ET质量浓度,大鼠脑组织5-HT及DA质量浓度有关。Objective To observe the effects of the compound Danggui Chuangge granule on NO,NOS,CGRP,ET and 5・HT and DA in plasma and brain tissue in migraine rats.Methods Sixty male Wistar rats were randomly divided into blank control group,model control group,low dose group(0.9 g/kg),middle dose group(1.8 g/kg),high dose group(3.6 g/kg)and sumaptan succinate group(0.05 g/kg),10 rats in each group.Rats in each group were administered continuously for 7 days.Thirty minutes after the last administration,except the blank control group,the rats in the other groups were given subcutaneous injection of nitroglycerin at the dose ratio of 10 mg/kg to body weight,and the rats in the blank control group were given subcutaneous injection of saline of equal volume at the hip.After 180 minutes of model establishment,rats in each group were anesthetized by abdominal cavity,plasma was separated,plasma NO,NOS,CGRP and ET were measured,brain tissue was dissected,tissue homogenate was prepared,and 5-HT and DA contents in brain tissue were measured.Results The middle and high dose of compound Danggui Chuangge granule could decrease the contents of NO, NOS, CGRP and ET in plasma and increase thecontents of 5-HT and DA in brain tissue of migraine rats. Conclusion The compound Danggui Chuangge granule haspreventive eff ect on migraine induced by nitroglycerin in rats. Its mechanism may be related to the regulation of NO,NOS, CGRP and ET contents in plasma and 5-HT and DA contents in brain tissue of rats.
