摘　　要：目的探索HCC临床标本中环指和WD重复结构域相关蛋白3(ring finger and WD repeat domain 3,RFWD3)的表达情况,并结合临床资料分析其与HCC预后的相关性。方法采用RT-PCR、Western blotting和免疫组化(IHC)法检测40例HCC患者癌组织及癌旁正常肝脏组织中RFWD3的mRNA及蛋白表达量,并运用GEPIA数据库分析RFWD3的表达和生存情况作为补充验证,同时结合临床资料分析其与HCC预后之间的相关性。结果RT-PCR、Western blotting和IHC检测发现,与癌旁组织相比,HCC组织中RFWD3的表达显著升高(P<0.05)。临床资料分析发现,RFWD3的高表达与HCC肿瘤大小显著相关(P<0.05)。生存分析发现,RFWD3高表达的HCC患者拥有更低的总体生存率和无瘤生存期(P<0.05)。结论RFWD3基因的高表达参与了HCC的发生和发展,可能是HCC一个重要的不良预后因素。Objective To explore the expression of ring finger and WD repeat domain 3(RFWD3)in HCC tissue samples,and to analyze its prognostic correlation with HCC based on clinical data.Methods Real-time polymerase chain reaction(RT-PCR),Western blotting and immunohistochemistry(IHC)were used to detect the m RNA and protein expression of RFWD3 in 40 cases of HCC samples and adjacent tissues.The GEPIA database was used as a supplementary verification for RFWD3 expression and survival analysis.The clinical relationship between RFWD3 and HCC was analyzed based on clinical data.Results The results of RT-PCR,Western blotting and IHC showed that the expression of RFWD3 in HCC tissues was significantly higher than those in adjacent tissues(P<0.05).Based on clinical data analysis,high expression of RFWD3 was markedly associated with tumor volume(P<0.05).Survival analysis found that patients with high expression of RFWD3 had a shorter overall survival(P<0.05)and disease-free survival(P<0.05).Conclusion High expression of RFWD3 is involved in the occurrence and development of HCC,which may be an important poor prognostic factor for HCC.
