摘  要:目的:探讨丹皮酚对宫颈癌细胞增殖的影响以及核因子E2相关因子2(Nrf2)和血红素加氧酶(HO-1)的作用。方法:用丹皮酚或DMSO处理Hela细胞后,MTT法、TUNEL检测细胞活性。采用免疫化学方法和westernblot检测Nrf2和HO-1信号的分子表达。结果:与对照组相比,丹皮酚处理的Hela细胞增殖活性降低,细胞凋亡指数升高,Nrf2、HO-1表达降低。结论:丹皮酚诱导Hela细胞凋亡,机制可能通过调节Nrf2与HO-1的表达。Objective: To investigate the effect of paeonol on proliferation of cervical cancer cells and the effect of nuclear factor E2 related factor 2(Nrf2) and Heme oxygenase factor 1(HO-1). Methods: After treatment of Hela cells with paeonol 400 mol/L or DMSO, MTT and terminal deoxynucleotidyl transferase end-labeling(TUNEL) method to detect cell proliferation activity. The molecular expression changes of Nrf2 and HO-1 signals were detected by immunohistochemistry and immunoblotting. Results: Compared with the control group, the proliferation of Hela cells treated with paeonol was decreased, and the apoptosis index was increased in paeonol group by TUNEL staining. The results of immunocytochemistry and western blot showed that the expression of Nrf2 and HO-1 in paeonol group was decreased. Conclusion: Paeonol can induce Hela cell apoptosis through regulating the expression of Nrf2 and HO-1.
