摘  要:目的构建一种富氧温控型骨组织工程材料并对其性能进行评价。方法本实验制备出含5%和10%1-溴十七氟辛烷的温控型骨组织工程材料,检测2种材料的凝固时间、pH值、释氧曲线,并通过筛选较优1-溴十七氟辛烷浓度的材料,检测该材料的微观结构、毒性及生物相容性。结果2种1-溴十七氟辛烷浓度的材料均可在37℃条件下凝固成胶,时间为4.5 min。2种材料的凝固时间和pH值比较差异无统计学意义(P>0.05)。2种材料浸提液中的氧分压均呈上升趋势,并在7 d时达到峰值,10 d内均可持续释放氧气,但1-溴十七氟辛烷浓度为10%的材料的释氧效能更高(P<0.01)。1-溴十七氟辛烷浓度为10%的温控型骨组织工程材料呈三维立体网状结构,孔隙率较高,且无细胞毒性,生物相容性良好,在体内可被完全降解。结论1-溴十七氟辛烷浓度为10%的温控型骨组织工程材料具有良好的释氧性能、良好的生物相容性和可降解性,可作为改善移植后早期低氧环境的缓释骨移植替代物。Objective To fabricate one kind of oxygen-enriched temperature-sensitive materials for bone tissue engineering and to further evaluate its performance.Methods In the experiments,5%and 10%perflubron of temperature-sensitive materials for bone tissue engineering were established.Solidification times,pH values and curves of oxygen releasing of the two kinds of materials were detected.The better concentration material of perflubron was selected to detect microstructure,toxicity and biocompatibility of the material.Results The two kinds of materials with different concentrations of perflubron were all solidified in 4.5 minutes at 37℃,and there were no significant differences in solidification time and pH values between two kinds of materisls(P>0.05).Levels of oxygen partial pressure in leaching liquor of two kinds of materials showed upward trend,and all reached a peak at the 7 th d,and oxygen was continuously released within 10 d in two kinds of materials.However the efficiency of oxygen releasing was higher in 10%perflubron than that in 5%perflubron(P<0.01).The 10%perflubron temperature-sensitive material for bone tissue engineering with the higher efficiency of oxygen releasing were the three-dimensional reticular formation,and it had higher interval porosity,no cytotoxicity with good biocompatibility,and it could be completely degraded in vivo.Conclusion The 10%perflubron temperature-sensitive material for bone tissue engineering has good efficiency of oxygen releasing,good biocompatibility and degradability,which may be used as surrogate of slow-release bone transplantation for improving hypoxia micro-environment after transplantation.
