摘 要:目的研究磷脂酶Cε1(PLCE1)蛋白在食管鳞状细胞癌(ESCC)及非肿瘤食管组织中的表达情况,探讨其与ESCC发病机制的关系。方法收集潮州市中心医院病理科存档的ESCC患者手术切除标本及非肿瘤患者内镜活检食管组织的蜡块。采用免疫组织化学染色PV-9000二步法(非生物素)检测非肿瘤患者食管正常鳞状上皮标本49例(A组)、肿瘤患者癌旁正常鳞状上皮标本73例(B组)和ESCC标本113例(C组)。结果 PLCE1蛋白在A组的表达水平明显高于C组和B组(P<0.05),且呈递减趋势。B组与C组之间的表达差异无统计学意义(P>0.05)。结论 PLCE1蛋白在A、B、C组中的表达差异,提示它可能参与ESCC发生,且其在正常食管鳞状上皮中的表达高于ESCC组织,提示其低表达在ESCC的发生过程中可能起到促进作用。Objective To investigate the expression of phospholipase C epsilon 1(PLCE1)protein in esophageal squamous cell carcinoma(ESCC)and non-tumor esophageal tissues and explore its relationship with the pathogenesis of ESCC.Methods Wax blocks of ESCC patients′surgically removed specimens and esophageal tissues of non-tumor patients were collected from the archives of pathology department of Chaozhou Centre Hospital.Expression of PLCE1 protein was determined by PV-9000 two-step method(non-biotin)of immunohistochemical(IHC)technique in 49 cases of normal esophageal squamous epithelium from nontumor cases(group A),73 cases of normal squamous epithelium adjacent to ESCC(group B)and 113 cases of ESCC(group C).Results The expression of PLCE1 protein in group A was significantly higher than that in group C and B,(both P<0.01),and showed a decreasing trend.There was no significant difference in the expression of PLCE1 protein between group B and C(P >0.05).Conclusion The difference expression of PLCE1 between group A,B and C suggests that PLCE1 protein may be involved in the pathogenesis of ESCC.The low expression of PLCE1 in group A may predict that PLCE1 protein may be a promoter gene of ESCC.
