摘 要:以忻州本地黄连为原料,采用回流法和超声法提取黄连中的小檗碱,得到的提取物与药店购买的复方黄连素片进行对比,研究其对枯草芽孢杆菌、大肠杆菌、变形杆菌的抑菌效果。结果表明,采用以乙醇为提取溶剂的回流法提取的黄连小檗碱的得率可达76%以上,超声波法提取的得率达50%;将3种生物碱液加到3种菌液体积分别为100 mL、200 mL、300 mL中共培养,在24 h、48 h、72 h后的菌落下降的数量呈上升趋势;在添加生物碱液24 h内,3种菌液下降的菌落数量均随着菌液体积的上升而增加,且黄连提取液的抑菌效果强于复方黄连素片。变形杆菌菌液体积为300 mL时,回流法的抑菌效果明显高于超声波法提取液和复方黄连素溶液。由此表明提取黄连小檗碱采用回流法效果更好。并且本试验表明忻州本地黄连中小檗碱的抑菌效果显著高于复方黄连素片(p=0.05)。本研究旨在优化黄连中小檗碱的提取工艺,以期为临床科学用药和制药等方面提供理论依据。The extraction of berberine from Coptis chinensis by reflux extraction and ultrasonic extraction was used to extract berberine from Coptis chinensis in Xinzhou.The extract was compared with the compound berberine tablet purchased by the drugstore to study the bacteriostasis effect on Bacillus subtilis,Escherichia coli and Proteus.The results showed that:the extraction rate of berberine from Coptis chinensis reached more than 76%by using ethanol as extraction solvent,and the extraction rate of berberberine from Coptis chinensis can reach 50%by ultrasonic method.When three different alkaloid extraction solutions were added to the three bacterial liquids with the product of 100 mL,200 mL and 300 mL respectively,the number of bacterial colonies decreased after 24 h,48 h and 72 h showed an upward trend.Within 24 h of administration,the number of colonies decreased by 3 strains of bacteria increased with the increase of bacteria volume no matter which inhibitory solution,and the bacteriostatic effect of the two extracts was stronger than that of compound berberberine tablets.When the liquid volume of proteobacterium was 300 mL,the bacteriostatic effect of reflux method was significantly higher than that of ultrasonic extraction solution and compound berberberine solution.The results preliminarily showed that it is better to extract berberine from Coptis chinensis by reflux method.The bacteriostatic effect of berberine in Xinzhou Coptis chinensis was significantly higher than that of compound berberberine tablets(p=0.05).This study provides scientific evidence for clinical scientific medicine and pharmaceuticals.
