摘 要:本文采用锐孔凝固浴法和复凝聚法复合法,以海藻酸钠和壳聚糖为壁材,将嗜酸乳杆菌、双歧杆菌和饲料包埋起来制备成益生菌微胶囊饲料,并对其制备工艺条件进行了研究。本试验将包埋率作为评价益生菌微胶囊饲料的主要指标,同时测定其粒径大小、悬浮率和溶解率,通过单因素试验和正交试验,研究了不同因素对这些理化性质的影响。结果表明:最佳制备工艺条件为海藻酸钠浓度1.25%,菌胶比例1∶10,氯化钙浓度1.5%,固化时间15 min,壳聚糖浓度1.5%。在这个工艺条件下,制备的益生菌微胶囊饲料粒径大小均匀,在1.60~2.00 mm;包埋率达到96.68%;而且在水中浸泡12 h后,悬浮率为77.96%,溶解率为13.04%;在模拟人工胃液中处理120 min后,仍然有5.2 lg(CFU/mL)的活菌数;在模拟人工肠液中处理120 min后,所包埋的活菌可全部释放出来,具备较好的漂浮性、水稳定性、耐酸性以及肠溶性。In this paper,sodium alginate and chitosan were used as shell-materials,the Lactobacillus acidophilus,Bifidobacterium and feed were coated to preparing probiotic microcapsules feed by perforation-curing with complex coacervation composite method,and the preparation conditions were studied.In this experiment,the encapsulation rate was used as the main index to evaluate the probiotic microcapsule feed,and its particle size,suspension rate and solubility rate were also determined,and the effects of different factors on these physical and chemical properties were studied by single factor test and orthogonal test.The results showed that the optimum preparation process conditions were determined as:the sodium alginate concentration 1.25%,the proportion of probiotics and sodium alginate 1∶10,calcium chloride concentration 1.5%,curing time 15 min,and chitosan concentration 1.5%.Under these conditions,the particle size of probiotic microcapsule feed is uniform,ranging from 1.60 to 2.00 mm;and the encapsulation rate is reached 96.68%.Moreover,the suspension rate is 77.96% and the solubility is 13.04% after soaking in water for 12 h;and still have a survival rate of 5.2 lg(CFU/mL)after 120 min in simulated artificial gastric juice;and the embedded live bacteria can be completely released after 120 min in simulated artificial intestinal fluid,which has good floatability,water stability,acid resistance and enteric solubility.
