摘  要:目的探讨RNA干扰ATP-柠檬酸裂解酶(ACLY)表达对结肠癌SW480细胞周期和凋亡的影响及其机制。方法将体外培养的SW480细胞分为Ctrl组、NC-siRNA组和ACLY-siRNA组,采用Western blot检测各组细胞中ACLY蛋白和蛋白激酶B(AKT)信号通路相关蛋白细胞周期蛋白D1(CyclinD1)、B细胞淋巴瘤/白血病-2(Bcl-2)、Bcl-2相关X蛋白(Bax)的表达以及AKT鳞酸化水平,噻唑蓝(MTT)法检测细胞存活率,流式细胞仪检测细胞周期分布和凋亡率。结果与Ctrl组相比,ACLYsiRNA组细胞中ACLY、CyclinD1、Bcl-2蛋白的表达水平、细胞存活率、细胞在S期和G2/M期百分比以及AKT鳞酸化水平均明显降低,而细胞在G0/G1期百分比、细胞凋亡率和Bax蛋白的表达水平均明显升高(P<0.05);NC-siRNA组与Ctrl组相比,差异无统计学意义(P>0.05)。结论RNA干扰ACLY表达可诱导结肠癌细胞周期阻滞和凋亡,其作用机制可能与抑制AKT信号通路的活化有关。Objective To investigate the effect of RNA interference ATP citrate lyase(ACLY)expression on cell cycle and apoptosis of colon cancer SW480 cells and its mechanism.Methods SW480 cells cultured in vitro were divided into Ctrl group(normal culture),NC-siRNA group(transfected NC-siRNA)and ACLY-siRNA group(ACLY-siRNA).The expression of ACLY protein and AKT signal pathway related proteins Cyclin D1,B cell lymphoma/lewkmia-2(Bcl-2),Bcl-2 Associated X Protein(Bax)and the level of protein kinase B(AKT)squamous acidification were detected by Western blot.cell viability was detected by MTT assay,cell cycle distribution and apoptosis rate were detected by flow cytometry.Results Compared with Ctrl group,the expression levels of ACLY,Cyclin D1,Bcl-2 protein,cell survival rate,cell percentage in S and G2/M phases and AKT squamous acidification level in ACLY-siRNA group were significantly lower,while the percentage of cells in G0/G1 phase,apoptotic rate and Bax protein expression level were significantly higher(P<0.05).But there was no significant difference between NA group and Ctrl group(P>0.05).Conclusion RNA interference ACLY expression can induce cell cycle arrest and apoptosis in colon cancer cells,and its mechanism may be related to inhibition of AKT signaling pathway activation.
