消瘤汤对裸鼠结直肠移植癌组织中促红细胞生成素肝细胞激酶B4及其配体EphrinB2表达的影响

摘　　要：目的探讨消瘤汤对裸鼠结直肠移植癌组织中促红细胞生成素肝细胞激酶B4(EphB4)及其配体EphrinB2蛋白表达的影响。方法将200只裸小鼠随机分为对照组、中药组、单抗组、联合组,各50只。另取25只裸鼠,将结直肠癌患者新鲜手术标本移植到培养裸鼠皮下;肿瘤成长后,取出肿瘤组织移植到各组裸鼠结肠系膜侧。移植3 d后,对照组给予蒸馏水灌胃,中药组给予消瘤汤灌胃,单抗组静脉注射贝伐单抗,联合组给予消瘤汤灌胃联合静脉注射贝伐单抗。给药6周后,检测各组裸鼠结直肠癌组织中EphrinB2和EphB4蛋白的表达情况。结果中药组、单抗组、联合组裸鼠结直肠癌组织中EphrinB2、EphB4的蛋白表达水平均低于对照组,且中药组和联合组均低于单抗组(均P<0.05),但中药组与联合组差异无统计学意义(P>0.05)。结论消瘤汤能抑制结直肠癌组织中EphrinB2和EphB4蛋白的表达,EphB4及其配体EphrinB2或可成为今后治疗结直肠癌的新靶点。Objective To explore the effect of Xiaoliu Decoction on the expressions of erythropoietin-producing hepatocyte kinase B4(EphB4)and its ligand EphrinB2 proteins in the cancer tissues of the colorectal transplantation tumor-bearing nude mice.Methods A total of 200 nude mice were randomly divided into control group,Chinese medicine group,monoclonal antibody group and combination group,with 50 cases in each group.Another 25 mice were selected,then the fresh surgical specimens from colorectal cancer patients were transplanted subcutaneously into the nude mice;after tumor growth,the tumor tissues were removed and then transplanted into the mesangium side of colon in the nude mice of each group.After 3 days of transplant,the control group and Chinese medicine group received intragastric administration of distilled and Xiaoliu Decoction respectively,while the monoclonal antibody group received intravenous injection of bevacizumab,and the combination group received intragastric administration of Xiaoliu Decoction combined with intravenous injection of bevacizumab.After 6 weeks of administration,the expressions of EphrinB2 and EphB4 proteins in the colorectal cancer tissues of nude mice were detected in each group.Results The expression levels of EphrinB2 and EphB4 proteins in the colorectal cancer tissues of nude mice were lower in the Chinese medicine group,the monoclonal antibody group and the combination group than those in the control group,and the level in the Chinese medicine group or the combination group were lower than that in the monoclonal antibody group(all P<0.05),but there was no statistically significant difference between the Chinese medicine group and the combination group(P>0.05).Conclusion Xiaoliu Decoction can inhibit the expressions of EphrinB2 and EphB4 proteins in the colorectal cancer tissues;EphB4 and its ligand EphrinB2 probably become the new therapeutic targets for colorectal cancer in the future.
