摘　　要：目的观察Rho GTP酶激活蛋白12(Rho GTPase-activating protein 12,ARHGAP12)在戊四氮(pentylenetetrazol,PTZ)致癫小鼠模型中的表达情况。方法25只C57BL/6小鼠随机分为癫痫组和对照组。分别应用Westernblot和免疫组织化学染色检测ARHGAP12的表达情况;应用免疫荧光染色对ARHGAP12进行定位。结果ARHGAP12定位于神经元的细胞膜;在癫痫模型中,ARHGAP12的表达水平较对照组明显增高(*P<0.05,**P<0.01)。结论ARHGAP12定位于海马神经元的细胞膜;在小鼠PTZ癫痫模型中,ARHGAP12高表达,提示其可能参与癫痫的形成。Objective To observe the expression of Rho GTPase-activating protein 12(ARHGAP12)in a mouse model of pentylenetetrazol(PTZ)-induced epilepsy.Methods Twenty-five C57 BL/6 mice were randomly divided into epilepsy group and control group.The expression of ARHGAP12 was analyzed by Western blot and immunohistochemical staining respectively.The expression of ARHGAP12 was detected by immunofluorescence stainin.Results ARHGAP12 was expressed at the cell membrane of neurons.In the epilepsy model,the expression level of ARHGAP12 was significantly increased than that of the control group(*P<0.05,**P<0.01).Conclusion ARHGAP12 was expressed at the cell membrane of hippocampal neurons.In the mouse PTZ epilepsy model,ARHGAP12 was highly expressed,suggesting that it may be involved in the formation of epilepsy.
