摘  要:目的通过多种生物信息学相关工具,对甲基转移酶样(METTL3)基因及蛋白进行结构及功能方面的分析。方法利用NCBI、STRING、Protscale等数据库或软件对METTL3的结构、功能、理化性质、生物活性等做出预测分析。结果METTL3基因编码一种不稳定的亲水性蛋白,该蛋白不具有信号肽结构及跨膜结构,主要分布于细胞核及胞质内。METTL3蛋白在人体内多种正常组织及器官中均有较为广泛的分布,主要参与核算甲基化及调节基因表达等生物学过程,同时也在肿瘤的发生发展及迁袭等生物学过程中起着重要的作用。METTL3具有特殊的三级结构,且拥有多个蛋白翻译后修饰位点。由METTL3突变所引起的肿瘤原发病例中,原发于乳房的病例所占比例较大。结论经系统地分析及预测,对METTL3基因有了更为深入和全面的了解,为该领域的研究提供了可靠的理论基础和研究思路。Objective To perform a systematic and informative analysis on structure and function of METTL3 gene and protein using related bioinformatic tools.Methods The structure,function,physico-chemical properties,and biological activity of METTL3 were predicted using NCBI,STRING,Protscale databases or softwares.Results The METTL3 gene was found to encode an unstable hydrophilic protein which does not possessany signal peptide structureor transmembrane structure,and is mainly localized in nucleus and cytoplasm.METTL3 protein is widely distributed in a variety of normal human tissues and organs,mainly involved in biological processes such as methylation and regulation of gene expression,and also play an important role in the development,progression and migration of tumors.METTL3 exhibits a special tertiary structure and has multiple post-translational modification sites.Among the cases of primary tumor caused by METTL3 mutation,proportion of primary breast tumors appears relatively high.Conclusion Systematic analysis and prediction offers a deeper and comprehensive understanding of METTL3 gene,and provides clues for anti-tumor studies with METTL3.
