摘  要:[目的]对广西贺州市荸荠种植区感染秆枯病的荸荠样本进行病原分离鉴定及分析病原菌的侵染过程,为荸荠秆枯病的有效防治提供参考。[方法]以常规组织分离法对发病明显的荸荠茎秆组织进行分离,采用传统的形态学对病原菌进行鉴定。通过制备分生孢子悬浮液接种于载玻片上培养,测定分生孢子萌发情况。利用病菌孢子悬浮液对荸荠茎秆进行离体接种,检测病菌的侵染过程。[结果]分离获得1株病原分离物(Ceh),经形态学鉴定该病原为荸荠柱盘孢菌(Cylindrosporium eleocharidis Lentz)。病菌分生孢子接种于载玻片后4 h开始萌发,8 h后大量萌发,12 h后形成大量附着孢,菌丝24 h后形成。病菌接种荸荠茎秆4 h后,分生孢子开始萌发并产生芽管;8~12 h芽管伸长,形成附着孢侵入荸荠茎秆表皮组织;24 h至7 d菌丝形成;接种8 d后,病菌在茎秆表面形成分生孢子盘。[结论]引起广西贺州市荸荠种植区荸荠秆枯病病原为荸荠柱盘孢菌(Cylindrosporium eleocharidis Lentz)。荸荠秆枯病菌分生孢子萌发和侵染过程可用于病原菌和寄主互作机理研究。[Objective]Pathogen causing stem blight of Chinese water chestnut in Hezhou production zone in Guangxi was identified and infection process of casual organism was analyzed to provide scientific references for protection of Chinese water chestnut planting.[Method]Isolation of pathogen was carried out from the diseased tissues of stem of Chinese water chestnut,and identification of isolate was performed by morphological characteristics.Conidia suspension was prepared to inoculate on slides to test conidia germination and to inoculate on stem of Chinese water chestnut in vitro to analyze infection process of pathogen,respectively.[Result]One isolate strain named Ceh was obtained and identified as Cylindrosporium eleocharidis Lentz with its morphological characteristics.The conidia started to germinate after 4 h incubation and reached the peak of germination at 8 h incubation,whereas a large number of appressoria were formed after 12 h incubation.Hyphae development was observed after 24 h incubation.The conidia began to germinate after 4 h inoculation and produced germ tubes on stem.The germinal tubes could invade any tissues on the epidermis of the stem and form appressoria after 8 h to 12 h inoculation.Hyphae was formed to infected the host cells within 7 days and acervuli were developed on the stem surface after 8 days inoculation.[Conclusion]Stem blight of Chinese water chestnut was regard as causing by Cylindrosporium eleocharidis in Hezhou Chinese water chestnut growing area in Guangxi.Analysis of conidia germination and infection process of casual organism could be used for the study interaction between pathogen and host.
