摘  要:目的观察芎芷地龙汤不同时间预防给药对偏头痛动物模型CREB1a、CREB1b mRNA表达的影响。方法将健康雄性SD大鼠36只,随机分为对照组(6只)、模型组(6只)、舒马普坦组(6只)、芎芷地龙汤1 d预防给药组(6只)、芎芷地龙汤3 d预防给药组(6只)、芎芷地龙汤7 d预防给药组(6只),按照预定给药,造模完成后2 h取材,用实时荧光定量聚合酶链式反应(real-time PCR)法测定三叉神经节、三叉神经脊束核、丘脑和硬脑膜CREB1a、CREB1b mRNA表达情况。结果与对照组比较,模型组大鼠CREB1a mRNA在三叉神经节、三叉神经脊束核表达水平明显降低(P<0.01),在硬脑膜和丘脑表达水平无明显变化(P>0.05)。给药干预后,三叉神经节、三叉神经脊束核CREB1a mRNA表达水平降低,以芎芷地龙汤7 d预防给药组和舒马普坦组降低明显(P<0.05);各组硬脑膜和丘脑CREB1a mRNA表达比较差异无统计学意义(P>0.05)。与对照组比较,模型组CREB1b mRNA在三叉神经节和三叉神经脊束核表达水平明显降低(P<0.05),在硬脑膜和丘脑表达水平无明显变化(P>0.05)。给药干预后,CREB1b mRNA在三叉神经节和三叉神经脊束核表达水平明显升高,以芎芷地龙汤7 d预防给药组和舒马普坦组增高明显(P<0.05);各组硬脑膜和丘脑表达比较差异无统计学意义(P>0.05)。结论芎芷地龙汤预防给药可以减少硝酸甘油诱发的三叉神经节、三叉神经脊束核CREB1a mRNA表达,可以增加三叉神经节、三叉神经脊束核CREB1b mRNA表达,芎芷地龙汤预防给药7 d效果好于预防给药3 d、1 d。Objective To investigate the effects of Xiongzhi Dilong decoction(XDD)preadministration at different time on the expression of cAMP response element binding protein(CREB)1a and CREB1b mRNA in migraine animal models.Methods Thirty-six healthy male Sprague-Dawley(SD)rats were randomly divided into six groups:normal saline group(n=6),migraine model group(n=6),sumatriptan group(n=6),1-day XDD preadministration group(n=6),3-day XDD preadministration group(n=6),7-day XDD preadministration group(n=6).The rats were administered according to the predetermined administration.The samples were collected after 2 hours of modeling.The expression of CREB1a and CREB1b mRNA in trigeminal ganglion,spinal trigeminal nucleus,thalamus,and dura mater were determined by the real-time polymerase chain reaction(PCR).Results Compared with the normal saline group,the expression of CREB1a mRNA in the trigeminal ganglion and spinal trigeminal nucleus was significantly increased in the model group(P<0.01).After drug intervention,the expressions of CREB1a mRNA in trigeminal ganglia and spinal trigeminal nucleus were decreased,which were significantly reduced in the 7-day XDD preadministration group and sumatriptan group(P<0.01).Compared with the normal saline group,the CREB1b mRNA expression in the trigeminal ganglia and the spinal trigeminal nucleus was significantly increased in the model group(P<0.01).After drug intervention,the expressions of CREB1b mRNA in trigeminal ganglia and spinal trigeminal nucleus were increased,which were significantly increased in the 7-day XDD preadministration group and sumatriptan group(P<0.01).Conclusion The preadministration of XDD can reduce the CREB1a mRNA expression in trigeminal ganglion and spinal trigeminal nucleus,and increase the CREB1b mRNA expression in trigeminal ganglion and spinal trigeminal nucleus which induced by nitroglycerin.The effect of 7-day XDD preadministration was better than that of 3-day or 1-day XDD preadministration.
