LncRNA HOTAIR在宫颈鳞癌组织中的表达及其与miRNA表达的相关性分析

摘　　要：目的探讨LncRNA HOTAIR在宫颈鳞癌组织中的表达情况,及其与宫颈鳞癌临床指标及miRNA表达的相关性。方法收集2015年3月一2018年3月在新疆医科大学附属肿瘤医院因宫颈鳞癌进行手术的宫颈组织标本(IB1-IIA2期)53例作为病例组,因子宫肌瘤或子宫腺肌症在新疆医科大学附属肿瘤医院进行全子宫切除术患者的正常宫颈组织46例作为对照组。采用Trizol法提取总RNA,利用qRT-PCR检测组织标本中LncRNA HOTAIR和4个miRNA的表达情况。2组间的LncRNA HOTAIR的比较采用student检验,2组间LncRNA HOTAIR与miRNA表达之间的相关性采用Spearman相关性分析。结果LncRNA HOTAIR在病例组标本中的表达量较对照组分值低(P=0.041);2组肿瘤临床分期(P=0.010)、淋巴结转移情况(P=0.035)、深层间质浸润深度(P=0.010)及分化程度(P=0.050)等指标的相关性具有统计学意义;宫颈鳞癌组织标本的LncRNA HOTAIR表达与miR-34a-5p(r=-0.477,P=0.003),miR-218-5p(r=-0.492,P=0.002)及miR-214-5p G=-0.376,P=0.026)的表达呈负相关。结枪LncRNA HOTAIR在宫颈鳞癌中组织表达异常,可能参与宫颈鳞癌的发生、发展,LncRNA HOTAIR有望成为宫颈癌早期诊断的生物学指标。Objective The aim of the study was to explore the expression of LncRNAHOTAIR in cervical squamouscell carcinoma and the statistical relationships between LncRNA HOTAIR expression and clinical features as well as the expression of the miRNAs.Methods Samples were collected from patients during March 2015 to March 2018, Affiliated Tumor Hospital of Xinjiang Medical University. Samples from patientswith cervical squamous cancer (IB1-IIA2) were considered to be case group, and normal cervicalsamples from patients with uterus myoma of uterus or adenomyosis, and performed total hysterectomywere considered to be control group・ Total RNA was extracted by Trizol method, and the expression ofLncRNA HOTAIR and 4 miRNAs in tissue samples was detected by qrt-PCR. The comparison of LncRNAHOTAIR between the two groups was conducted by student test, and the correlation between LncRNAHOTAIR and miRNA expression between the two groups was analyzed by Spearman correlation analysis.Student's t test was used to analyze the expression differences between case and control group. Spearmancorrelation analysis was performed to analyze the relationships between LncRNA HOTAIR expression andmiRNA expression. Results LncRNA HOTAIR expression was decreased in uterine cervical squamouscellcarcinoma tissue compared to control group (P =0.041), and correlation with clinical stage (P =0.010),lymph node metastasis (P =0.035), the depth of deep interstitial invasion (P =0.010) and differentiation(P =0.050) has statistical significance. Correlation analysis reveal that LncRNA HOTAIR expression wasnegatively correlated with the expression of miR-34a-5p (厂= -0.477,P =0.003), miR-218-5p (厂=0.492,P =0.002) and miR-214-5p (厂=—0.376=0.026) in cervical cancer. Conclusion The expression of LncRNAHOTAIR was aberrant in cervical cancer and was first discovered to be related to clinical indicator.The LncRNA HOTAIR might be used as biomarkers in early diagnosis of cervical cancer.
