摘 要:目的:观察银杏黄酮对非酒精性脂肪肝大鼠肝脏组织中脂质和炎症因子水平的影响。方法:大鼠分为正常对照组,模型组和银杏黄酮低、中、高剂量组,每组9只。第1~4周,正常对照组给予正常饲料,其他4组给予高脂饲料以构建非酒精性脂肪肝模型;第5~10周,银杏黄酮低、中、高剂量组大鼠以80、160、320 mg/kg的银杏黄酮灌胃处理,1次/d,其他2组给予生理盐水。第10周末,采集血清,检测ALT、AST水平。摘取肝脏,计算肝脏指数,用生化分析仪检测肝组织中TG、LDL-C、FFA、SOD、MDA水平,Western blot法检测肝组织中IL-1β、TNF-α蛋白的表达水平。结果:与正常对照组比,模型组大鼠肝脏指数升高,血清ALT、AST水平升高,肝组织中TG、LDL-C、FFA水平升高,IL-1β、TNF-α蛋白表达水平升高,MDA水平升高,SOD活性下降(P<0.05)。银杏黄酮低、中、高剂量组以上指标均较模型组改善,且随剂量升高,改善更明显(P<0.05)。结论:银杏黄酮能够降低非酒精性脂肪肝大鼠脂质和炎症因子水平,减轻肝损伤。Aim:To explore the effects of ginkgo flavone on lipid and inflammation factor level in the liver of rats with nonalcoholic fatty liver disease.Methods:The experiment included normal control group,model group,low dose group,middle dose group and high dose group,9 rats in each group.During the first 4 weeks,rats in the latter 4 groups took high-fat diet,and those in normal control group took normal diet.From 5 to 10 weeks,rats in the low,middle and high dose groups were treated with 80,160 and 320 mg/kg ginkgo flavone once a day.At the end of the 10th week,the liver index was calculated.The serum levels of ALT and AST were determined.The levels of TG,LDL-C,FFA,SOD and MDA in liver tissue were detected by biochemical analyzer.The expressions of IL-1βand TNF-αin liver tissue were detected by Western blot.Results:Compared with normal control group,liver index in model group increased,serum ALT and AST increased,the levels of TG,LDL-C and FFA in liver tissue increased,the expression levels of IL-1βand TNF-αprotein increased,MDA level increased and SOD activity decreased(P<0.05).Compared with model group,the above indexes all improved in the low,middle and high dose groups,especially in the high dose group(P<0.05).Conclusion:Ginkgo flavone could improve lipid and inflammation factor level in rats with nonalcoholic fatty liver disease.
