摘 要:目的:探讨SPOCK2基因表达上调对前列腺癌细胞增殖的影响。方法:通过Real-time PCR及Western Blot检测前列腺癌组织及细胞中SPOCK2基因的表达。进一步通过基因重组技术上调前列腺癌细胞系DU145及LNCaP中SPOCK2基因表达,通过CCK8检测细胞增殖情况变化,通过流式细胞仪检测细胞周期及凋亡情况。结果:前列腺癌组织中SPOCK2表达显著低于正常对照组(P<0.05),应用基因重组片段有效上调SPOCK2基因表达后,转染组细胞增殖率均显著低于阴性对照组及空白对照组,差异有显著统计学意义(P<0.05)。转染组G0/G1期细胞百分比显著高于阴性对照组及空白对照组,差异有显著统计学意义(P<0.05)。转染组S期显著低于阴性对照组及空白对照组,差异有显著统计学意义(P<0.05)。DU145细胞转染组凋亡率显著高于阴性对照组及空白对照组,差异有显著统计学意义(P<0.05)。结论:SPOCK2上调可以抑制细胞增殖,促进细胞凋亡,使细胞停滞于G0/G1期,表明该基因可能通过影响细胞增殖,作为抑癌基因在前列腺癌的发生发展中发挥重要作用。Objective:To investigate the effect of SPOCK2 gene up-regulation on the proliferation of the prostate cancer cells.Methods:Real-time PCR and Western Blot was used to detect the expression of SPOCK2 in the prostate cancer tissues and cells.The expression of SPOCK2 gene in prostate cancer cell lines DU145 and LNCaP was upregulated through gene recombination.Cell proliferation was detected by CCK8 assay,and the cell cycle and apoptosis were detected by flow cytometry.Results:The expression of SPOCK2 was significantly lower in prostate cancer tissues than that in prostate hyperplasia tissues(P<0.05).The expression of SPOCK2 gene was effectively upregulated by gene recombination,and the proliferation rate of the transfected group was significantly lower than that of the negative control group and the vector control group(P<0.05).The percentage of G 0/G 1 phase cells in the transfection group was significantly higher than that in the negative control group and the vector control group(P<0.05).The S phase of the transfection group was significantly lower than that of the negative control group and the vector control group(P<0.05).The apoptosis rate of DU145 cells of transfected group was significantly higher than that of the negative control group and the vector control group(P<0.05).Conclusion:Up-regulation of SPOCK2 can inhibit cell proliferation,promote apoptosis,and arrest cells in G 0/G 1.SPOCK2 may play an important role in the development of prostate cancer by affecting cell proliferation.
