摘　　要：目的探讨缺氧诱导因子-1α(HIF-1α)和HIF-2α蛋白的表达情况及与宫颈癌患者临床特征及预后的关系。方法应用免疫组化法检测68例宫颈癌患者的活检组织标本和30例宫颈良性病变中HIF-1α、2α蛋白的表达情况,并进行对比,分析它们表达情况与宫颈癌临床病理特征及预后的关系。结果免疫组化结果显示,宫颈癌组织标本中HIF-1α和HIF-2α蛋白的阳性表达率分别为61.76%和70.59%,明显高于良性病变组(均P<0.01);两者的阳性表达与淋巴结转移及放疗前贫血均相关(均P<0.05),而HIF-2α蛋白表达与肿瘤分期相关(P<0.05)。生存分析结果提示,HIF-1α蛋白阴性的宫颈癌患者的中位总生存期(OS)明显长于HIF-1α蛋白阳性者(P<0.05)。结论在宫颈癌患者中,HIF-1α和HIF-2α蛋白的表达显著升高,它们对宫颈癌患者预后有良好的预测价值,两者的高表达提示疾病可能预后不良。Objective To investigate the expression of hypoxia-inducible factor-1α(HIF-1α)and hypoxia-inducible factor-2α(HIF-2α)proteins in cervical carcinoma and its relationship with clinical feature and prognosis of patients.Methods The expression of HIF-1αand HIF-2αin 68 samples of cervical cancer and 30 samples of cervicitis tissues were detected by immunohistochemistry and the relationship of HIF-1αand HIF-2αexpression with clinicopathological features and prognosis of patients was analyzed.Results Immunohistochemistry showed that the positive expression rates of HIF-1αand 2αin cervical cancer tissues were 61.76%and 70.59%,which were significantly higher than those in cervicitis tissues(all P<0.01).There was a positive correlation between the high expression of HIF-1αand HIF-2αwith lymph node metastasis and anemia before radiotherapy(P<0.05),and HIF-2αexpression was positively correlated with tumor stage(P<0.05).The survival analysis showed that the median overall survival(OS)of HIF-1αnegative patients was significantly longer than that of HIF-1αpositive patients(P<0.05).Conclusion The expression of HIF-1αand HIF-2αis up-regulated in cervical cancer tissues,and patients with high expression of HIF-1αand HIF-2αmay suggest a poor prognosis.
