摘 要:为研究五大连池药泉湖沉积物中细菌多样性,采用16S rDNA高通量测序的方法对从药泉湖沉积物样品进行序列分析,发现样品中获的序列涵盖了40个菌门,丰度大于1%的菌门有变形杆菌门(Proteobacteria)、放线菌门(Actinobacteria)、拟杆菌门(Bacteroidetes)芽单胞菌门(Gemmatimonadetes)、疣微菌门(Verrucomicrobia)和绿菌门(Chlorobi)。优势菌门为Proteobacteria和Actinobacteria。此外,优势菌属为杆菌属(Bacterium),还包含丰度大于1%的属鞘氨醇单胞菌属(Sphingomonas)、嗜甲基菌属(Methylotenera)、芽单胞菌属(Gemmatimonas)、慢生根瘤菌(Bradyrhizobium)。其中,鞘脂杆菌属(Muiclaginibacter)为T01位点独有的属,在药泉湖不同位点的样品细菌群落表现出一定的差异。上述结果表明五大连池药泉湖沉积物中具有丰富的细菌多样性,并且与其生存的环境紧密相关。16S rDNA high-throughput sequencing method was used to sequence the sediment samples of Yaoquan lake in order to study the bacterial diversity in the sediments of Yaoquan lake in wudalianchi lake.The sequences obtained from the sediments of Yaoquan lake covered 40 phylum and Proteobacteria(26.29%),Actinobacteria(27.76%),Bacteroidetes(12.82%),Gemmatimonadetes(4.16%),Verrucomicrobia(3.10%),Chlorobi(2.24%)were more than 1% abundant of phylum.And Sphingomonas(17.21%),Methylotenera(6.72%),Gemmatimonas(1.35%),Bradyrhizobium(1.02%)were more than 1% abundant of genus.Among them,Mucliaginibacter(9.87%)was the exclusive genus of T01 locus.The bacterial communities of sediment samples collected at different sites in Yaoquan lake were different to some extent.The above results indicated that the sediment of Yaoquan lake in wudalianchi was rich in bacterial diversity and closely related to its living environment.
